Pharmacology of HIV integrase inhibitors [PDF]

open access: yesCurrent Opinion in HIV and AIDS, 2012
The purpose of this paper is to review recent and relevant pharmacology data for three HIV integrase inhibitors: raltegravir (marketed), dolutegravir and elvitegravir (both in Phase III drug development).

HIV-1 integrase modulates the interaction of the HIV-1 cellular cofactor LEDGF/p75 with chromatin [PDF]

open access: yesRetrovirology, 2011
Background Chromatin binding plays a central role in the molecular mechanism of LEDGF/p75 in HIV-1 DNA integration. Conflicting results have been reported in regards to the relevance of the LEDGF/p75 chromatin binding element PWWP domain in its HIV-1 ...

Novel therapeutic strategies targeting HIV integrase

open access: yesBMC Medicine, 2012
Integration of the viral genome into host cell chromatin is a pivotal and unique step in the replication cycle of retroviruses, including HIV. Inhibiting HIV replication by specifically blocking the viral integrase enzyme that mediates this step is an ...

Integrase variability and susceptibility to HIV integrase inhibitors: impact of subtypes, antiretroviral experience and duration of HIV infection [PDF]

open access: yesJournal of Antimicrobial Chemotherapy, 2009
Little is known about the extent and predictors of polymorphisms potentially influencing the susceptibility to HIV integrase inhibitors (INIs).Genetic sequences of HIV integrase were obtained from INI-naive patients at two European clinics. The 39 amino acid changes at 29 integrase positions so far associated with INI resistance were examined according

Mutations Located outside the Integrase Gene Can Confer Resistance to HIV-1 Integrase Strand Transfer Inhibitors

open access: yesmBio, 2017
Resistance to the integrase strand transfer inhibitors raltegravir and elvitegravir is often due to well-identified mutations in the integrase gene. However, the situation is less clear for patients who fail dolutegravir treatment.

HIV-1 Integrase and ALLINI Structure.

open access: yes, 2023
A. Domain Map of HIV-1 Integrase. B. The preclinical lead ALLINI BI-224436. Chemical features common to the ALLINI class are highlighted, including a carboxylic acid moiety (salmon), a tert-butoxy moiety (green), and a large hydrophobic appendage at the ...

Emerging patterns in HIV integrase resistance. [PDF]

open access: yesAIDS
We assessed integrase resistance in 837 treatment-experienced people with HIV (PWH) with virological failure (2022–2024) in Portugal. Major resistance mutations were found in 5.5%, with N155H and R263K being the most common. Resistance was more frequent in non-B subtypes and often co-occurred with resistance to other antiretroviral classes.

Integrase and integration: biochemical activities of HIV-1 integrase [PDF]

open access: yesRetrovirology, 2008
Integration of retroviral DNA is an obligatory step of retrovirus replication because proviral DNA is the template for productive infection. Integrase, a retroviral enzyme, catalyses integration. The process of integration can be divided into two sequential reactions.

Molecular monitoring of HIV-1 drug resistance in Ifakara HIV-1 Cohort, Tanzania [PDF]

open access: yes, 2013
HIV-1 resistance is one of the problems affecting success of antiretroviral therapy programmes worldwide. Many studies on efficacy of ART programmes, specifically on drug resistance, have been conducted in developed countries but not in developing ...
