TPI1 Loss Triggers a Metabolite‐Driven Mitochondrial Redox Vulnerability via the SARM1–cADPR–Ca2+ Axis

open access: yesAdvanced Science, EarlyView.
Metabolite‐driven redox stress governs cancer cell senescence. TPI1 deficiency elevates DHAP, initiating SARM1‐dependent cADPR‐Ca2+ release. Mitochondrial ROS surge subsequently induces DNA damage and senescence, offering a conserved therapeutic target in multiple cancers.

G‐Quadruplexes: Structural Diversity and Emerging Roles in Biomolecular Condensation

open access: yesAdvanced Science, EarlyView.
G‐quadruplexes (G4s) fold into diverse intra‐ and intermolecular structures, positioning them as emerging regulators of biomolecular condensation. Mechanistically, G4s autonomously form condensates, act as structural platforms to initiate and stimulate condensation, or induce phase transitions.

Npro of classical swine fever virus enhances HMGB1 acetylation and its degradation by lysosomes to evade from HMGB1-mediated antiviral immunity

open access: yesVirus Research
Classical swine fever virus (CSFV) can dampen the host innate immunity by destabilizing IRF3 upon its binding with viral Npro. High mobility group box 1 (HMGB1), a non-histone nuclear protein, has diverse functions, including inflammation, innate ...

A Supramolecular Thermal Switch for Precision Pyroptosis via Host−Guest Recognition and Electrostatic Interactions

open access: yesAdvanced Science, EarlyView.
A supramolecular pyroptotic thermal switch encapsulates I3− within β‐cyclodextrin (β‐CD) and undergoes CaCO3 mineralization. Upon lysosomal acidification, CaCO3 is degraded, releasing Ca2+ and I3−. Concurrently, BODIPY activation converts I3− into I2, inducing oxidative stress and Ca2+ overload, which together trigger caspase‐3/GSDME‐mediated ...

HMGB1 protein: regulation by specific E3 ubiquitin ligases

open access: yesGenes & Cells
HMGB1 belongs to the high mobility group (HMG) protein family, characterized by the presence of structurally conserved DNA-binding HMGB domain. In addition to two DNA-binding domains connected by a short linker, HMGB1 contains a short N-terminal sequence and an intrinsically disordered C-terminal region.

Spatiotemporally Ultrasound‐Controlled Nanoparticles Reprogramming Immunostimulatory Antigen‐Presenting Cancer‐Associated Fibroblasts to Enhance Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
We developed an ultrasound‐controlled biomimetic nanoplatform, mRNA/V@M NPs, to co‐deliver CD74 mRNA and V‐9302 for fibrotic TNBC therapy. CD74 mRNA reprograms myCAFs into antigen‐presenting apCAF cells through the CD74‐MHC II pathway, while V‐9302 induces ICD and activates MHC I‐CD8+ T cell immunity.

In Situ Profiling of Nanoscale Strains Uncovers Mechano‐Architectural Predictors of Aging and Osteoarthritis Emergence

open access: yesAdvanced Science, EarlyView.
In situ mechanical loading of osteoarthritis‐prone murine knee joints reveals that spatial alterations in subchondral plate microarchitecture precede cartilage loss and promote a shift from coordinated epiphyseal strain dissipation in healthy joints to focal subchondral strain concentration in osteoarthritis‐prone joints.

Engineering‐Modulated Molybdenum Enzymes Strategy for Tumor‐Specific Metabolic‐Immunotherapy

open access: yesAdvanced Science, EarlyView.
Biodegradable molybdenum sulfide nanoparticles were synthesized via a one‐pot strategy to increase molybdenum enzyme activity and thus effectively potentiate anti‐tumor immunity by integrating molybdenum‐based metalloimmunotherapy with hydrogen sulfide gas therapy.

HMGB1 B-Box Domain Associates Promote Protein–Polyelectrolyte Interactions

open access: yesThe Journal of Physical Chemistry B
HMGB1, a nuclear DNA-binding protein, can be secreted by activated immune cells or passively released from damaged cells. In such cases, HMGB1 functions as an alarmin that activates the immune system. Excessive inflammation may lead to pathogenesis, whereas this response can be dampened by polyanion binding, which impedes further receptor recognition ...

Hypothalamic Control of Liver Health and Disease: From Circuits to Pathophysiology and Therapies

open access: yesAdvanced Science, EarlyView.
This review delineates the hypothalamic circuits that control liver homeostasis via autonomic and neuroendocrine pathways. Dysregulation of this hypothalamus–liver axis drives disease progression across a spectrum including steatotic liver disease, liver inflammation and injury, fibrosis, cirrhosis, and hepatocellular carcinoma.
