HPAEC–PAD analysis of oligogalacturonic acids in strawberry juice

Food Chemistry, 1999
Abstract High performance anion-exchange chromatography (HPAEC), coupled with pulsed amperometric detection (PAD), was used to identify and quantify oligogalacturonic acids (OGAs) in strawberry juice. This method allowed good separation and detection of OGAs with degree of polymerization (DP) ® MAX enzyme, for 2 h at 45°C, produced juice with by ...

Fast automated online xylanase activity assay using HPAEC-PAD

Analytical and Bioanalytical Chemistry, 2017
In contrast to biochemical reactions, which are often carried out under automatic control and maintained overnight, the automation of chemical analysis is usually neglected. Samples are either analyzed in a rudimentary fashion using in situ techniques, or aliquots are withdrawn and stored to facilitate more precise offline measurements, which can ...

Quantification of sugars in wheat flours with an HPAEC-PAD method

Food Chemistry, 2015
An HPAEC-PAD method has been developed and validated for the simultaneous determination and quantification of six sugars (glucose, isomaltose, maltose, maltotriose, maltotetraose and maltopentaose) in wheat flours, by extraction with water and precipitation of proteins with Carrez II.

N-Glycosylation Analysis by HPAEC-PAD and Mass Spectrometry

2013
Changes in protein glycosylation are a hallmark of most types of cancer including ovarian carcinoma. The structural elucidation of glycans is technically challenging and it requires complementary chromatographic and spectroscopic techniques among others. Here, we describe the profiling of N-glycans from glycoproteins of SKOV3 ovarian carcinoma cells by

HPAEC-PAD analysis of monoclonal antibody glycosylation

1995
Publisher Summary Monosaccharide and oligosaccharide analysis using HPAEC-PAD are two of the methods biopharmaceutical manufacturers use to characterize glycan structures and to monitor the lot-to-lot consistency of therapeutic glycoproteins. This chapter describes the use of these methods in conjunction with oligosaccharide standards and endo- and ...

Molecular size distribution assessment of Haemophilus influenzae vaccine containing lactose by HPAEC-PAD and colorimetric assays

Analytical Biochemistry, 2022
Molecular size distribution of Haemophilus influenzae type b (Hib) conjugate vaccine is an important indicator for its immunogenicity and stability. Molecular size distribution was evaluated by High-Performance Protein Chromatography on Sepharose CL-4B column, and fractions were pooled. The use of high flow rate, incorporation of a calibration standard

Analysis of Pectin Structure by HPAEC-PAD

1996
Pectins are complex, branched, acidic polysaccharides that are located throughout the plant primary cell wall matrix. Pectin structure in cell walls has been described as block-like (Jarvis 1984) in which smooth and hairy regions exist (de Vries et al. 1982). The smooth pectin regions consist of homogalacturonan (McNeil et al. 1984).

HPAEC-PAD for oligosaccharide analysis—novel insights into analyte sensitivity and response stability

Analytical and Bioanalytical Chemistry, 2017
The rising importance of accurately detecting oligosaccharides in biomass hydrolyzates or as ingredients in food, such as in beverages and infant milk products, demands for the availability of tools to sensitively analyze the broad range of available oligosaccharides.

HPAEC-PAD oligosaccharide analysis to detect adulterations of honey with sugar syrups

Food Chemistry, 2008
A method previously validated in our laboratory to study the oligosaccharide profile of honey was used in this work to detect adulterations of corn syrups (CS) and high fructose corn syrups (HFCS) in genuine honey samples. High molecular weight oligosaccharides (DP3-DP16) of 9 sugar syrups and 25 honey samples were analysed by HPAEC-PAD.
