Purification and preparation of Marchantia polymorpha Auxin Response Factor 2 for phase separation studies

open access: yesFEBS Open Bio, EarlyView.
We describe detailed protocols for the purification and preparation of Marchantia polymorpha Auxin Response Factor 2 (MpARF2). This protein is fused to an MBP solubility tag and an mNG fluorescent tag and is purified from Escherichia coli. The presented procedures make it possible to study MpARF2 assemblies, which could arise from phase separation ...

The suppression of Antarctic bottom water formation by melting ice shelves in Prydz Bay

open access: yesNature Communications, 2016
Antarctic bottom water (AABW) production is critical to the global ocean overturning circulation. Here, the authors show new observations of AABW formation from seal CTD data in Prydz Bay, East Antarctica that highlights its susceptibility to increased ...

Retrogressive thaw slump susceptibility in the northern hemisphere permafrost region

open access: yesEarth Surface Processes and Landforms
Mean annual temperatures in the Arctic and subarctic have increased in recent decades, increasing the number of permafrost hazards. Retrogressive thaw slumps (RTSs), triggered by the thawing of ground ice in permafrost soil, have become more common in ...

Protocol for quantifying miRNA trafficking across the endosomal membrane

open access: yesFEBS Open Bio, EarlyView.
An in vitro protocol measures miRNA uptake into endosomes isolated from mammalian cell extracts, which are free of subcellular contaminants. Performed at 37 °C in the presence of ATP, it ensures the import of single‐stranded miRNA into the endosomal lumen.

Impact of surface melt and brine infiltration on fracture toughness of ice shelves [PDF]

open access: yesThe Cryosphere
Ice shelves are heterogeneous composites of firn, meteoric ice, refrozen melt and brine-saturated ice. The properties and distribution of these elements control ice shelf response to stress and susceptibility to fracturing.
