B‐Cell Repertoire of Children With Atopic Dermatitis Exhibits Altered IgE Maturation Associated With Allergic Food Sensitization

IgH repertoire sequencing of a large atopic dermatitis cohort reveal reduced IgE somatic hypermutation and increased IgE diversity associated with food allergen sensitization. Presence of a dog during the first year of life is associated with increased IgE somatic hypermutation, possibly countering the effect of food sensitization.

Long‐Term Effect of Allergen Immunotherapy in Responder Local Allergic Rhinitis Patients: Symptom Control, and Prevention of Asthma and Allergic Sensitizations

LAR patients who respond to AIT during the first year and are treated for two more years continue improving (less symptoms and medication intake) during the following seven years. AIT prevents the onset of asthma and allergic sensitizations in responder LAR individuals during the seven years following discontinuation.

Decreased Severity of Oral Immunotherapy Induced Gastrointestinal Eosinophilic Reactions Following Revisions in Milk‐Treatment Protocol

A revised milk OIT protocol with lower daily allergen load and slower dose escalation reduced the severity and delayed onset of OIT‐induced gastrointestinal eosinophilic reactions (OITIGER). Elevated baseline absolute eosinophil count (AEC > 600/μL) was a consistent independent risk factor for OITIGER, while older age was protective.

Oral Immunotherapy Induces Shift in lncRNA Expression Modulating Allergen‐Specific Immune Responses

We used PBMC transcriptome from 50 hen's egg‐allergic patients to study the role of lncRNA in OIT. Positive correlation was found between 17 lncRNAs and the IL‐4/IL‐13 pathway in vivo, and the expression of the selected lncRNA aligned with CXCL8 upon PBMC egg‐restimulation.

Basophil Activation Test as Biomarker of Severity and Threshold of Allergic Reactions to Cow's Milk During Oral Food Challenges

The threshold of reactivity during fresh milk challenges was 70% lower than that of baked milk challenges, reflecting the higher allergenicity of fresh milk. BAT was the only biomarker statistically significantly different between severity and threshold groups, for baked milk and fresh milk allergies, respectively.
