An implantable hydrogel is designed to hold gene transfection agents engineered to turn early recurrent tumor cells into generators of synthetic EVs. These synthetic EVs can express engineered miR‐26a (E‐miR‐26a) for highly sensitive detection and PD‐1 (a PD‐L1‐blocking agent) for therapeutic intervention, thereby enabling early detection and ...

Immune checkpoint inhibitors for brain metastases
PURPOSE OF REVIEW: Metastasis of cancer to the brain typically portends a poor prognosis and often results in significant morbidity, including from the side effects of treatment. More effective therapies for patients with brain metastases are needed. The

ABSTRACT While targeted therapies have improved outcomes in lung adenocarcinoma (LUAD), many patients still lack targetable mutations. Here, we identified alpha‐L‐fucosidase 2 (FUCA2) as a crucial driver of LUAD by preventing cellular senescence. Mechanistically, through the restriction of fucosyltransferase 3 (FUT3)‐mediated α‐1,3‐fucosylation of ...

A case series of immune checkpoint inhibitor-induced gastritis
Immune checkpoint inhibitors (ICIs) have become a cornerstone in the treatment of various solid tumors due to their proven efficacy. However, their use is associated with immune-related adverse events (irAEs), which arise from the immune response ...

Glioblastoma (GBM) exhibits RFX7 silencing via promoter hypermethylation, which downregulates PIK3IP1 to activate PI3K/AKT signaling, increase lactate accumulation, and enhance histone H4K12 lactylation. This lactylation upregulates PD‐L1/CSF1 expression and drives the immunosuppressive microenvironment formation.

ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.
