Nudix Hydrolase 13 Impairs the Initiation of Colorectal Cancer by Inhibiting PKM1 ADP‐Ribosylation

open access: yesAdvanced Science, Volume 12, Issue 13, April 3, 2025.
This study identified NUDT13 as a roadblock in the progression from colorectal adenoma to CRC, and revealed that NUDT13 stabilizes PKM1 protein by reducing the PARylation that is catalyzed by PARP1, thereby reprogramming the metabolic propensity of CRC cells.

Muscle‐Derived Small Extracellular Vesicles Mediate Exercise‐Induced Cognitive Protection in Chronic Cerebral Hypoperfusion

open access: yesAdvanced Science, EarlyView.
sEVs have a critical role in orchestrating interorgan crosstalk and mediating exercise‐induced therapeutic effects. Lin et al. demonstrates that sEVs miR‐17/20a‐5p mediates the muscle‐brain crosstalk and emphasizes the central role of mTOR signaling in executing molecular programs that can protect brain health in response to exercise. Abstract Physical

NAT10 Promotes Gastric Cancer Liver Metastasis by Modulation of M2 Macrophage Polarization and Metastatic Tumor Cell Hepatic Adhesion

open access: yesAdvanced Science, Volume 12, Issue 15, April 17, 2025.
The N‐acetyltransferase‐10 (NAT10)‐mediated mRNA N4‐acetylcytidine modification enhances the stability of CXCL2 and KLF5 mRNA. Then, secreted CXCL2 promotes M2 macrophage polarization and oncostatin M secretion, activating NAT10 expression via STAT3 signaling.

Conjugated Lithocholic Acid Activates Hepatic TGR5 to Promote Lipotoxicity and MASLD‐MASH Transition by Disrupting Carnitine Biosynthesis

open access: yesAdvanced Science, EarlyView.
Takeda G protein‐coupled receptor 5 (TGR5) serves as a central regulator in the metabolic dysfunction‐associated steatotic liver disease (MASLD)/metabolic dysfunction‐associated steatohepatitis (MASH) transition via conjugated lithocholic acid (LCA). Mechanistically, TGR5 orchestrates lipotoxicity‐induced cell death through its interaction with CD36 ...

HNF3α Targets Nckap1l and Promotes Renal Fibrosis Following Ischemia‐Reperfusion Injury

open access: yesAdvanced Science, EarlyView.
Hepatocyte Nuclear Factor 3 alpha (HNF3α) is significantly upregulated in renal tubular epithelial cells of CKD patients and fibrotic mice. Deletion of HNF3α mitigates IRI‐induced renal fibrosis, while overexpression worsens it. HNF3α directly regulates NCK‐associated protein 1‐like (Nckap1l), whose overexpression exacerbates fibrosis.
