The mast cell stabilizing activity of Chaga mushroom critical for its therapeutic effect on food allergy is derived from inotodiol

International Immunopharmacology, 2018
While an anti-allergic effect of Chaga mushroom (Inonotus obliquus) has been indicated, its therapeutic effect on allergy and immunoregulatory mechanisms and chemical constituents directly responsible for that are hardly known. We examined the effect of 70% ethanol extract of Chaga mushroom (EE) and its dichloromethane (DF) and aqueous (AF) fractions ...

CE9A215 (inotodiol), a lanostane-type oxysterol, mitigates LPS-induced sepsis through multifaceted mechanisms

European Journal of Pharmacology
Dysregulated host response against infection triggers sepsis that leads to multiple organ dysfunction due to uncontrolled inflammatory responses. Despite marked progress in understanding of sepsis, numerous clinical trials for treatment of sepsis have proven daunting and a new therapeutic approach is highly needed.

Structure determination of inonotsuoxides A and B and in vivo anti-tumor promoting activity of inotodiol from the sclerotia of Inonotus obliquus

Bioorganic & Medicinal Chemistry, 2007
Two new lanostane-type triterpenoids, inonotsuoxides A (1) and B (2) along with three known lanostane-type triterpenoids, inotodiol (3), trametenolic acid (4), and lanosterol (5), were isolated from the sclerotia of Inonotus obliquus (Pers.: Fr.) (Japanese name: Kabanoanakake) (Russian name: Chaga).

Rapid Isolation and Purification of Inotodiol and Trametenolic Acid from Inonotus obliquus by High‐speed Counter‐current Chromatography with Evaporative Light Scatting Detection

Phytochemical Analysis, 2011
AbstractIntroductionIn Eastern Europe, especially Russia, the fruiting body of Inonotus obliquus has been used as a folk medicine for cancer since the sixteenth or seventeenth century. Inotodiol and trametenolic acid are considered to be the main bioactive compounds of the fruiting body of the mushroom.

Glucose fed-batch integrated dissolved oxygen control strategy enhanced polysaccharide, total triterpenoids and inotodiol production in fermentation of a newly isolated Inonotus obliquus strain

Process Biochemistry, 2018
Abstract Submerged fermentation is promising for producing secondary metabolites of Inonotus obliquus, a folk medicine to treat cancer and other diseases. However, there limited progress work in strain selection or process optimization in order to enhance production of its bioactive metabolites.

Inotodiol, a lanostane triterpenoid, from Inonotus obliquus inhibits cell proliferation through caspase-3-dependent apoptosis.

Anticancer research, 2008
To investigate the antitumor effect of Inonotus obliquus Pilat, the antiproliferative effect of lanostane triterpenoids from a chloroform extract of I. obliquus sclerotia against mouse leukemia P388 cells was assessed.Cell viability was measured by MTT assay. Caspase-3/7 activity and DNA fragmentation were evaluated to analyze apoptosis induction.

Inonotus obliquus (Chaga Fungus): In Vitro Culture and the Production of Inotodiol, Sterols, and Other Secondary Metabolites

1994
The wood-rotting fungus, Inonotus obliquus (Pers. ex Fr.) Pilat, Hymenochaetaceae, Basidiomycetes, is found in northern hardwood forests, chiefly in the circumpolar or mountainous areas of Europe, Asia, and North America. The fungus occurs widely in birches (Betula), but it is occasionally found in alder (Alnus), ash (Fraxinus) and rowan (Sorbus).
