Transposon Insertional Mutagenesis Models of Cancer

Cold Spring Harbor Protocols, 2014
Transposon-based insertional mutagenesis in the mouse provides a powerful approach for identifying new cancer genes. Transposon insertions in cancer genes are selected during tumor development because of their positive effect on tumor growth, and the transposon insertion sites in tumors thus serve as tags for identifying new cancer genes.

Insertional mutagenesis: Transposon-insertion libraries as mutagens in yeast

2002
Publisher Summary Transposon-based insertional libraries are exceptionally useful as a means of screening for desired phenotypes and expression patterns. As compared to mutations generated by classical methods of chemical treatment and ultraviolet irradiation, transposon insertions are easy to identify: the transposon-encoded sequence serves as a ...

Activation Tagging and Insertional Mutagenesis in Barley

2010
The process of activation tagging in plants involves the random distribution of plant regulatory sequences throughout the genome. The insertion of a regulatory sequence in the vicinity of an endogenous gene can alter the transcriptional pattern of this gene resulting in a mutant phenotype that arises from excess functional gene product.

Saturation Mutagenesis by Codon Cassette Insertion

Cold Spring Harbor Protocols, 2019
Saturation mutagenesis by cassette insertion introduces a library of site-specific changes into a specific DNA sequence within a target gene and is especially useful for analyzing the effect of specific residues on the structure and function of a protein.

Tn10 insertional mutagenesis in Pasteurella multocida

Veterinary Microbiology, 1996
The objective of this study was to identify a transposon that will randomly and stably integrate in the genome of Pasteurella multocida. Using the suicide conjugative delivery system, pLOF, containing a kanamycin resistance marked Tn10 insertion element, we determined that Tn10 is effective in producing insertional mutations in this species.

Controlling P element insertional mutagenesis

Trends in Genetics, 1988
Abstract Two new methods for insertional mutagenesis in Drosophila are now available. They are based on the ability to control experimentally the number of stable P element insertions per mutagenized strain. One method produces about 10 insertions per genome and has the potential to replace hybrid dysgenesis as a means for cloning genes by ...

Tnt1 Insertional Mutagenesis in Medicago truncatula

2018
Legumes play irreplaceable roles in sustainable agriculture due to their unique capability of fixing gaseous nitrogen in the atmosphere and turning into plant-usable ammonium through interaction with rhizobia. With the completion of genome sequencing of several model and non-model legumes, it is highly desirable to generate mutant populations for ...

Insertional mutagenesis in Arabidopsis thaliana

Plant Science, 1991
Abstract Recently, a number of mutant gene loci in the Arabidopsis thaliana plant genome have been identified through insertional mutagenesis. In this review, we evaluate different methods used for Agrobacterium tumefaciens -mediated T-DNA insertional mutagenesis with regard to their mutation frequencies and conclude that a major breakthrough in ...

Insertional Mutagenesis in Mouse Mammary Tumorigenesis

1991
Over the past few years, several surprising discoveries have been made regarding cellular oncogenes which are activated in mouse mammary tumors. These genes provide a particularly good example of the theme of this volume: host cell oncogenes activated by insertion of proviral DNA.

[Integration of AAV vectors and insertional mutagenesis].

Medecine sciences : M/S, 2016
Recombinant AAV vectors (rAAV) are considered as very efficient tools for in vivo gene transfer. Accordingly, several preclinical and clinical gene therapy trials use these vectors to treat inherited and acquired diseases. rAAV vectors possess the capacity to persist for a long term in the transduced tissue in a transcriptionally active, extra ...
