Metabolomics: quantification of intracellular metabolite dynamics
Biomolecular Engineering, 2002The rational improvement of microbial strains for the production of primary and secondary metabolites ('metabolic engineering') requires a quantitative understanding of microbial metabolism. A process by which this information can be derived from dynamic fermentation experiments is presented. By applying a substrate pulse to a substrate-limited, steady

The emergence of intracellular metabolite signaling networks
IUBMB Life, 2016AbstractThe signals of intracellular metabolites are integrated into cellular signaling networks by forming metabolite‐derived posttranslational modifications or by non‐covalently binding to proteins. These signals serve to coordinate cell metabolism and to regulate various aspects of cell physiology.

Automated Sampling Device for Monitoring Intracellular Metabolite Dynamics
Analytical Biochemistry, 1999An automated sampling device coupled to a stirred tank reactor was developed for monitoring intracellular metabolite dynamics. Sample flasks fixed in transport magazines were moved by a step engine in a way that each sample flask was filled within 220 ms, resulting in a sampling rate of 4.5 s-1.

Intracellular metabolites of cisplatin in the rat kidney
Cancer Chemotherapy and Pharmacology, 1989The appearance of low-molecular-weight metabolites of cisplatin in the cytosol of cells from the cortex and outer medulla of the rat kidney has been examined using HPLC up to 24 h following cisplatin administration. Comparison was made between these metabolites and those present in plasma, urine and liver.

Extra- and intracellular metabolite concentrations for murine hybridoma cells
Applied Microbiology and Biotechnology, 1992Intra- and extracellular metabolite concentrations were determined for hybridoma cells grown in tissue culture flasks in batch culture. Significant differences were found for intracellular lactate and amino acid concentrations depending on the culture medium. AB2-143.2 cells cultivated in Dulbecco's modified Eagle's (DME) medium supplemented with 50 mM

RNA Sensors of Intracellular Metabolites
2012Gene expression in bacteria can be controlled through many different possible regulatory strategies. The “expression” of a given gene can be described as a function of all of the molecular processes involved in the conversion of the gene’s information first into messenger RNA (mRNA) transcripts and then into the corresponding proteins.

Artificial Caprolactam-Specific Riboswitch as an Intracellular Metabolite Sensor
ACS Synthetic Biology, 2019Caprolactam is a monomer used for the synthesis of nylon-6, and a recombinant microbial strain for biobased production of nylon-6 was recently developed. An intracellular biosensor for caprolactam can facilitate high-throughput metabolic engineering of recombinant microbial strains. Because of the mixed production of caprolactam and valerolactam in the

Reaction engineering methods to study intracellular metabolite concentrations
1996The analysis of intracellular metabolite concentrations is of basic importance for metabolic engineering of microorganisms. In vivo NMR-spectroscopy as a non-invasive technique to measure intracellular metabolite concentrations and rapid sampling devices as invasive techniques are reviewed. The methods are discussed from a reaction engineering point of

Quenching Methods for the Analysis of Intracellular Metabolites
2013Sampling and quenching methods for intracellular metabolite analysis in mammalian cells in adherent and suspension culture are described. Quenching of adherent cells is achieved by application of hot air after removal of the supernatant by suction.
