A Biomimetic Lubricant Captures Hyaluronic Acid In Situ to Regenerate Cartilage: From Bench to Bedside

open access: yesAdvanced Science, EarlyView.
(a) The outstanding lubricative properties of natural articular cartilage arise from a composite coating composed of HA, lubricin, and phospholipids at its surface. Notably, lubricin facilitates the enrichment of synovial HA onto cartilage surfaces, and HA complexes with phospholipids, a key component to maintain extremely low friction under high ...

Materials‐Guided Gene‐Ionizable Lipid Nanoparticles to Reverse Iron‐Associated Immune Resistance in Renal Cancer

open access: yesAdvanced Science, EarlyView.
ABSTRACT Iron overload is a common metabolic disturbance in cancer and contributes to poor outcomes in renal cell carcinoma (RCC), yet its effects on the tumour immune microenvironment remain unclear. Here we identify a previously unrecognized immunosuppressive axis in which iron overload downregulates the palmitoyltransferase ZDHHC12 in CD8+ T cells ...

Kinsenoside Targets IDH1 to Restore Microglial Immune‐Metabolic Homeostasis for Alzheimer's Disease Therapy

open access: yesAdvanced Science, EarlyView.
Dysregulated TCA cycle contributes to Alzheimer's disease (AD) pathogenesis. Here, we show that microglial isocitrate dehydrogenase 1 (IDH1) is a critical driver. Elevated IDH1 disrupts citrate metabolism and mitochondrial function, exacerbating AD pathology.

Secreted Nonstructural Protein 3 is a Pathogenic Determinant of Orbivirus

open access: yesAdvanced Science, EarlyView.
This study uncovers a conserved PIP2‐dependent secretory pathway of orbivirus NS3 that induces vascular leakage. Pharmacological disruption of PIP2‐NS3 interaction significantly reduces viral pathogenicity and provides protective efficacy in murine models, establishing PIP2‐mediated NS3 secretion as both a key virulence determinant and a promising ...

Using the amide proton signals of intracellular proteins and peptides to detect pH effects in MRI

Nature Medicine, 2003
In the past decade, it has become possible to use the nuclear (proton, 1H) signal of the hydrogen atoms in water for noninvasive assessment of functional and physiological parameters with magnetic resonance imaging (MRI). Here we show that it is possible to produce pH-sensitive MRI contrast by exploiting the exchange between the hydrogen atoms of water

Crystal structure of importin‐α bound to a peptide bearing the nuclear localisation signal from chloride intracellular channel protein 4

The FEBS Journal, 2011
It has been reported that a human chloride intracellular channel (CLIC) protein, CLIC4, translocates to the nucleus in response to cellular stress, facilitated by a putative CLIC4 nuclear localization signal (NLS). The CLIC4 NLS adopts an α‐helical structure in the native CLIC4 fold.
