Isolation of human and porcine islets of langerhans and islet transplantation in pigs
Journal of Surgical Research, 1974Abstract Using a modification of the collagenase digestion, Ficoll gradient separation technique, we isolated the islets of Langerhans from fresh human pancreata removed from braindead cadavers. Comparison of the tissue insulin/amylase ratios between whole pancreas and isolated material revealed a significant purification of islet from acinar tissue.

Abstract Twenty partially inbred German shepherd dogs were made diabetic by intravenous administration of streptozotocin (25 mg/kg) 1 and 3 days after partial pancreatectomy. Ten diabetic dogs received intraportal transplants of allogeneic islets of Langerhans isolated by collagenase digestion and Ficon gradient separation.

Effect of Antithymocyte Globulin on Islet of Langerhans Transplantation
Nephron, 1978Lewis rats were treated with streptozotocin to induce hyperglycemia and glycosuria (400-600 mg/dl). Transplantation of approximately 1,000 dissociated islets obtained from collagenase-treated pancreases from 4 donors will promptly correct induced diabetes.

As the preservation of the isolated islets is difficult, we attempted to keep these tissues viable by use of an organ culture. Islets of Langerhans from adult Wistar rats were isolated by a collagenase technique and cultured in air-CO2 (95-5%) incubator at 37 degrees C.

Insulin-releasing activity was studied for the purpose of clarifying the function of cryopreserved islets. Cryopreserved pancreatic islets were also transplanted into the portal veins of recipient rats.

Transplantation of isologous islets of langerhans in diabetic rats
Acta Diabetologica Latina, 1975Isologous isolated islets of Langerhans were transplanted into the peritoneum and, via the portal vein, into the liver of diabetic rats. In both groups almost normal blood glucose and serum insulin levels were achieved for a period of three months. Glucose tolerance tests were markedly improved.
