Immunohistochemistry as a tool for identifying PDGFRA amplification in central nervous system tumors. [PDF]

open access: yesBrain Pathol
PDGFRA gene amplification is now recognized as a diagnostically and prognostically relevant molecular alteration in central nervous system tumors. Herein, we demonstrated that PDGFRA immunohistochemistry is a highly specific and sensitive biomarker for identifying PDGFRA amplification and should be part of the neuropathologist's routine panel of ...

Clinical Impact of MGMT Promoter Methylation in IDH‐Mutant Gliomas: Influence of Threshold Selection and Clinical Confounding

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Background The clinical relevance of MGMT promoter methylation in IDH‐mutant gliomas remains controversial in the era of molecular classification. We aimed to systematically evaluate its clinical relevance by integrating quantitative assessment, cutoff exploration, and adjustment for clinical confounding.

Advanced Materials for Biologics Delivery to Brain Tumors

open access: yesAdvanced Materials, EarlyView.
Material innovation is central to unlocking the therapeutic potential of biologics against many central nervous system diseases, including brain cancer. By engineering carriers with controlled transport, targeting, and release properties, advanced materials can overcome the blood–brain barrier and tumor microenvironment, improving the delivery of ...

The Cancer Cell Metabolic Reprogramming Remodels the Tumor Microenvironment: Molecular Mechanisms and Therapeutic Strategies

open access: yesAdvanced Science, EarlyView.
This review elucidates how cancer cell metabolic reprogramming—across glucose, lipid, amino acid, and nucleotide pathways—remodels the tumor microenvironment to suppress anti‐tumor immunity and promote immune escape. Targeting these metabolic axes offers promising strategies to overcome immunotherapy resistance and enhance cancer treatment.

Targeting the KAT7/H3K14ac/RAC2 Axis Mediates OXPHOS to Promote Stemness Maintenance and Malignancy in Glioblastoma

open access: yesAdvanced Science, EarlyView.
SOX2 upregulates KAT7 in glioma stem cells, where KAT7 deposits H3K14 acetylation at the RAC2 promoter to activate RAC2 transcription. RAC2‐GTP subsequently promotes PAK1/2/3 phosphorylation, increasing tricarboxylic acid cycle flux and mitochondrial respiration to support glioblastoma malignancy.

Nasal Tuft Cell Markers Are Predictive of Overall Survival in Sinonasal Neuroendocrine and Sinonasal Undifferentiated Carcinomas

open access: yesInternational Forum of Allergy &Rhinology, EarlyView.
ABSTRACT Background Sinonasal neuroendocrine carcinoma (SNEC) and sinonasal undifferentiated carcinoma (SNUC) are rare aggressive malignancies with no established prognostic markers. Because features of POU2F3+ small cell lung carcinomas have been identified in both tumors, we evaluated immunostaining for POU2F3, a tuft cell transcription factor, and ...

From anthelmintic to neuro‐oncology: A systematic review of mebendazole repurposing for brain tumour therapy

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Aim Mebendazole (MBZ), a benzimidazole anthelmintic with established clinical use, has emerged as a repurposing candidate for primary brain tumours due to its multimodal anticancer actions and central nervous system penetrance. This systematic review synthesizes preclinical and clinical evidence evaluating MBZ's efficacy, mechanisms of action ...

Dose‐related propofol resistance in COVID‐19 patients: Altered drug plasma levels, mechanistic insights from neuronal‐derived extracellular vesicles and miRNA analysis

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Aim Critical care for COVID‐19 patients presents unique challenges, particularly in sedation management. Patients demonstrate notable dose‐related resistance to propofol, potentially due to complex pharmacokinetic alterations. Emerging evidence suggests that plasma extracellular vesicle‐associated miRNA represents valuable predictive ...

A nanoplatform that induces dual‐amino acid deprivation to reverse tumor immunosuppression and enhance metabolic immunotherapy

open access: yesBMEMat, EarlyView.
ZIF‐8‐based nanoparticles co‐delivering CB‐839 (glutaminase inhibitor) and 1‐MT (IDO1 inhibitor), dual‐targeting glutamine/tryptophan metabolism to induce immunogenic cell death, activate STING, block kynurenine production, reverse immunosuppression, and enhance cancer immunotherapy to suppress primary/distant tumors.
