Ramification Amplification: A Novel Isothermal DNA Amplification Method

Molecular Diagnosis, 2001
We have developed a novel isothermal DNA amplification method with an amplification mechanism quite different from conventional PCR. This method uses a specially designed circular probe (C-probe) in which the 3' and 5' ends are brought together in juxtaposition by hybridization to a target.

Detection of tetracycline using isothermal amplification

2017 IEEE 12th International Conference on Nano/Micro Engineered and Molecular Systems (NEMS), 2017
PCR is an important biosensing method since it can amplify DNA by many orders of magnitude. However, for food safety and environmental monitoring, the target molecule is usually not DNA. Here we demonstrate a detection method that integrates competitive assay format, magnetic bead separation, and isothermal DNA amplification for sensitive detection of ...

Quadratic isothermal amplification for the detection of microRNA

Nature Protocols, 2014
This protocol describes an isothermal amplification approach for ultrasensitive detection of specific microRNAs (miRNAs). It achieves this level of sensitivity through quadratic amplification of the target oligonucleotide by using a Bst DNA polymerase-induced strand-displacement reaction and a lambda exonuclease-aided recycling reaction.

Isothermal DNA amplification in vitro: the helicase-dependent amplification system

Cellular and Molecular Life Sciences, 2009
Since the development of polymerase chain reaction, amplification of nucleic acids has emerged as an elemental tool for molecular biology, genomics, and biotechnology. Amplification methods often use temperature cycling to exponentially amplify nucleic acids; however, isothermal amplification methods have also been developed, which do not require ...

[A method of isothermal gene amplification].

Comptes rendus de l'Academie des sciences. Serie III, Sciences de la vie, 1999
A gene amplification technique applied to the detection of HIV and which can be implemented without programmable thermocyclers is described. The reaction products are high molecular weight strands of DNA that can be detected using simple methods, even without electrophoresis. This is a promising technique for in situ amplification experiments.
