PCR ITS-RFLP for Penicillium Species and Other Genera

2016
Among numerous molecular methodologies developed for highly specific identification of filamentous fungi isolates, here we describe restriction digestion analysis of the ITS products as an easy method to identify isolates of filamentous fungi. This technique is a rapid and reliable method appropriate for routine identification of filamentous fungi ...

Multiple displacement amplification of DNA for ITS-RFLP analysis of individual juveniles of Bursaphelenchus

Nematology, 2005
AbstractDifferentiation of the plant-pathogenic pinewood nematode, Bursaphelenchus xylophilus, from non-pathogenic Bursaphelenchus species is difficult because of high morphological similarities among closely related species. In recent years, ITS-RFLP analysis has become a useful tool for Bursaphelenchus species identification.

ITS-RFLP and ITS sequence analysis of a foliar endophytic Phyllosticta from different tropical trees

Mycological Research, 2003
Different isolates of a foliar endophytic species of Phyllosticta were isolated from different tropical tree species in India to examine genetic variation among the isolates. Internal transcribed spacer-restriction fragment length polymorphism (ITS-RFLP) analysis did not detect any variation among the isolates, suggesting that they all belong to the ...

An ITS-RFLP method to identify black Aspergillus isolates responsible for OTA contamination in grapes and wine

International Journal of Food Microbiology, 2007
Ochratoxigenic mycobiota in grapes from representative wine regions in Valencia was identified. Black aspergilli were predominant among the different Aspergillus spp. isolated. Restriction digestion analysis of the ITS products was tested as a rapid method to identify isolates of black Aspergillus species from grapes. Restriction endonuclease digestion

ITS-RFLP and sequence analysis of endophytes from Acianthus, Caladenia and Pterostylis (Orchidaceae) in southeastern Queensland

Mycological Research, 2005
We used ITS-RFLP and sequence analysis to determine the identities of the fungal endophytes of six terrestrial orchid species from southeastern Queensland, a region previously unexplored in this context. Pure cultures of orchid--colonising fungi were obtained and fungal identities were assessed by means of ITS-PCR, RFLP analysis, sequence comparison ...

ITS-RFLP characterization of black Aspergillus isolates responsible for ochratoxin A contamination in cocoa beans

European Food Research and Technology, 2009
In the present study, ochratoxigenic mycobiota in cocoa beans was identified at species level by digestion of the ITS products using the endonucleases HhaI, NlaIII and RsaI. Of the 132 isolates of Aspergillus section Nigri collected from cocoa beans, 89 were identified as A. tubingensis, 27 as A. niger, 10 as A. tubingensis-like and 6 as A. carbonarius.

Genetic relationships of pathogenic Typhula species assessed by RAPD, ITS-RFLP and ITS sequencing

Mycological Research, 2000
Several species of Typhula cause diseases of cereals and grasses at low temperatures. Taxonomic separation of T. phacorrhiza, T. incarnata and T. ishikariensis was investigated using RAPD, ITS-RFLP and nucleotide sequencing of the ITS region. Using 10 RAPD primers on genomic DNA or eight restriction enzymes on PCR-amplified ITS regions, banding ...

ITS–RFLP and ITS sequence diversity in Pisolithus from central and eastern Australian sclerophyll forests

Mycological Research, 2001
Fifty-three isolates of Pisolithus were obtained from various locations in central and eastern Australia. These, along with two isolates from south-east Asia and one from USA, were compared using ITS–RFLP and ITS sequencing analyses. Results of the RFLP analysis initially grouped the isolates into eight RFLP types.

Ectomycorrhizal community composition of Pinus tabulaeformis assessed by ITS-RFLP and ITS sequences

Botany, 2010
Ectomycorrhizal (ECM) fungal composition was examined in a Pinus tabulaeformis Carr. forest. A total of 28 root samples of P. tabulaeformis were collected in June and September. Thirty-five ECM morphotypes were identified according to ECM morphological characters, and 26 ECM fungi were identified based on the analyses of ITS-RFLP and ITS sequences ...

ITS–RFLP- and RAPD-based genetic variability of Trypanosoma cruzi I, human and vector strains in Santander (Colombia)

Parasitology Research, 2009
Chagas disease is a severe public health problem in Latin-American countries. In Colombia, the predominance of Trypanosoma cruzi I has been described in the literature, with a broad heterogeneity between strains. However, most of the studies carried out centered on isoenzyme analysis, with a smaller number that focus on other molecular methods. In this
