Kainic acid derivatives with anticonvulsant activity

beta-Kainic acid, and the glycine and amino-methylphosphonate derivatives of alpha- and beta-kainic acid, have been injected intracerebroventricularly in DBA/2 mice, that show sound-induced seizure responses. An anticonvulsant effect is observed with marked protection against the tonic and clonic phases of the seizure response.

Distribution and persistence of kainic acid in brain

The distribution of 3H-kainic acid in rat brain was studied as a function of time after injections of 5 nmoles into the neostriatum, substantia nigra or cerebellum. More than half of the injected material had disappeared from the injection site and the brain by 1/2 hour post injection.

Abstract The neurotoxicity induced by intrastriatal injections of kainic acid was greater in rats drinking approximately 4 ml ethanol per day for 16 days before injection than in controls. Intraperitoneal injection of 5 mg/kg baclofen (parachlorophenyl GABA), 30 min before the kainic acid injections had a slight protective action.
