Genetic polymorphism of the caprine kappa casein gene

Polymorphism in the goat kappa casein gene was studied using the base excision sequence scanning (BESS) method and sequencing. Seven polymorphic sites, corresponding to single nucleotide transversions were detected. Three of these were silent mutations while the other four produced amino acid substitutions.

The carbohydrate content of kappa casein.

k-Casein which was electrophoretically (Starch gel) homogeneous was hydrolyzed with the enzyme Pronase P and the resultant hydrolyzate was fractionated by column chromatography. A fraction, rich in carbohydrate, was isolated by use of Sephadex G-25. Analysis showed that it contained at least nine different components.

The far-IR spectrum of kappa-casein, a giant molecular diode, has been obtained in the region 50–250 cm−1 and its electronic conduction properties have been determined. The conductivity is found to be four orders of magnitude higher in the ℱ-helix form than in a globule.

A method for kappa-casein genotyping of bulls.

A method for kappa-casein genotyping in bulls has been developed. By analysis of DNA polymorphisms we are able to discriminate between the kappa-casein variant A and B in the bulls. This method will be an efficient tool in selection for the most desirable kappa-casein variant.
