Optimizing the ER Cutoff in HER2‐Positive Breast Cancer: ER ≥ 50% Predicts Low pCR Rates and Resistance to Antibody‐Drug Conjugates in the Neoadjuvant Setting

open access: yesCancer Medicine, Volume 15, Issue 10, October 2026.
ABSTRACT Background Estrogen receptor (ER)‐positive/HER2‐positive breast cancer demonstrates significantly lower rates of pathological complete response (pCR) to neoadjuvant HER2‐targeted therapies compared to ER‐negative/HER2‐positive disease. However, the optimal ER positivity cutoff for clinically meaningful patient stratification remains undefined.

Pi3K- and Mtor-Dependent Mechanisms of Lapatinib Resistance and Resulting Therapeutic Opportunities [PDF]

open access: yes, 2014
Breast cancers with HER2 amplification represent 20-25% of breast cancer cases and are frequently responsive to the HER2 kinase inhibitor lapatinib, but generally for only short duration.

Development of acquired resistance to lapatinib may sensitise HER2-positive breast cancer cells to apoptosis induction by obatoclax and TRAIL

open access: yesBMC Cancer, 2018
Background Lapatinib has clinical efficacy in the treatment of trastuzumab-refractory HER2-positive breast cancer. However, a significant proportion of patients develop progressive disease due to acquired resistance to the drug.

Mapping Mutations and Signaling Network Interactions to Guide Precision Therapy in Gallbladder Cancer

open access: yesCancer Reports, Volume 9, Issue 10, October 2026.
ABSTRACT Background Gallbladder cancer (GBC) is a biologically complex malignancy arising from the epithelial lining of the gallbladder, with adenocarcinoma constituting the major histological subtype. Early detection is challenging because of vague clinical manifestations and the organ's deep‐seated anatomical location, resulting in diagnosis at ...

Lapatinib reduces hypertrophy in ErbB2 transgenic mice.

open access: yes, 2012
Wild type and ErbB2 transgenic male and female mice were treated with lapatinib via gavage for 3 weeks, starting at P10.5 (n = 5–12 per genotype per treatment group per sex).

Response and resistance to lapatinib in HER2-overexpressing breast cancer [PDF]

open access: yes, 2017
HER2 overexpression has been implicated in breast cancer. Lapatinib is a dual EGFR/HER2 tyrosine kinase inhibitor used in advanced HER2-positive breast cancer patients who have previously progressed on trastuzumab treatment.

Lapatinib in breast cancer

open access: yesAnnals of Oncology, 2007
Aberrant activation of some members of human epidermal growth factor receptor (HER) family plays a key role in breast carcinogenesis. Lapatinib is an oral dual tyrosine kinase inhibitor selective for inhibition of epidermal growth factor receptor (EGFR/ErbB1) and HER2/ErbB2.

Targeting the PI3K/Akt Signaling Pathway: A Promising Therapeutic Strategy to Reduce Cancer Mortality and Overcome Chemoresistance

open access: yesHealth Science Reports, Volume 9, Issue 10, October 2026.
ABSTRACT Background and Aims The phosphatidylinositol 3‐kinase (PI3K)/Akt signaling pathway is a central regulator of cellular properties that are essential for cancer progression, including cellular growth, survival, metabolism, and angiogenesis. Dysregulated PI3K/Akt signaling, often caused by genetic alterations in PIK3CA, loss of PTEN, or the ...

Lapatinib-induced enhancement of mitochondrial respiration in HER2-positive SK-BR-3 cells: mechanism revealed by analysis of proteomic but not transcriptomic data

open access: yesFrontiers in Molecular Biosciences
Dual inhibitors of HER2 and EGFR, such as lapatinib, have shown significant efficacy for the therapy of HER2-positive breast cancer. Previous experiments showed that in cell cultures, the efficacy of lapatinib was significantly reduced by exposure to ...
