A UV laser-scanning confocal microscope for the measurement of intracellular Ca2+

Cell Calcium, 1994
Modifications to the optics of a conventional confocal laser-scanning microscope were made to allow imaging intracellular Ca(2+)-dependent fluorescence with a UV laser (351 or 364 nm). Modifications included: (1) a chromatic compensation lens in the laser path; (2) the design of a practically achromatic relay lens; (3) a longer tube length for the ...

The Intermediate Optical System of Laser-scanning Confocal Microscopes

1990
This text explains some of the basics of intermediate optical systems in confocal microscopes. Important microscope components are discussed in more detail in other chapters: lasers (Chapter 5) and other light sources (Chapter 6), objective lenses (Chapter 7), special optical elements (Chapter 3), fibers (Chapter 26), and detectors (Chapter 12 ...

A custom confocal and two-photon digital laser scanning microscope

American Journal of Physiology-Heart and Circulatory Physiology, 2000
We describe a custom one-photon (confocal) and two-photon all-digital (photon counting) laser scanning microscope. The confocal component uses two avalanche photodiodes (APDs) as the fluorescence detector to achieve high sensitivity and to overcome the limited photon counting rate of a single APD (∼5 MHz).

Spherical-coordinate scanning confocal laser microscope

Optical Engineering, 1995
We present the design, construction, and characterization of a spherical-coordinate scanning, reflection-mode confocal laser microscope (SCSLM). Two system designs are presented, namely a conventional bulk optics object-scanning design and a preliminary optical-fiber-based two-axis optics/one-axis object scanning version.

Three-dimensional microscopy using a confocal laser scanning microscope

Optics Letters, 1985
In a scanning laser microscope detecting fluorescent light from the specimen, the depth-discriminating property of confocal scanning has been used to carry out optical slicing of a thick specimen. The recorded digital images constitute a three-dimensional raster covering a volume of the specimen.

A video rate confocal laser scanning microscope

Optical Society of America Annual Meeting, 1992
A video rate confocal laser scanning microscope (CLSM) for biological applications has been developed for manufacture. This microscope is used for viewing thin "optical sections" of a sample by rejecting the signal from out-of-focus planes. The microscope consists of a multiline argon ion laser, a galvanometer and acousto-optic deflector (AOD) for ...

Ultrastructural and confocal laser scanning microscopic examination of TUNEL‐positive cells

The Journal of Pathology, 1997
TdT-mediated dUTP-biotin nick end labelling (TUNEL) has been widely used for detecting cells with DNA fragmentation or apoptotic cells. However, since the concept of apoptosis is based on cellular ultrastructure, it is important to identify the morphological features of TUNEL-positive cells.

Scanning and detection techniques used in a confocal scanning laser microscope

Journal of Microscopy, 1990
SUMMARYA two‐mirror scanning mechanism for confocal microscopy is described. No optical components, in addition to the scanning mirrors, are used. Design criteria and performance of the scanner are discussed. The photometric linearity of a detector unit incorporating a photomultiplier tube is reported, and a dual detector unit with tunable split ...

Optically Corrected Diode Laser for Confocal Scanning Microscope

Frontiers in Optics 2007/Laser Science XXIII/Organic Materials and Devices for Displays and Energy Conversion, 2007
We propose the use of optically corrected laser diode in the confocal scanning microscope. The resulted beam has no astigmatism and produces a round diffraction limited spot on a sample.
