Comparative efficacy and safety of anakinra and canakinumab in patients with VEXAS syndrome – an international multicenter study

open access: yesArthritis &Rheumatology, Accepted Article.
Objectives The aim of this study was to compare differences in clinical response, drug survival, and adverse event rates between anakinra and canakinumab in VEXAS syndrome. Methods This multicenter international study includes VEXAS patients from France, Israel, and Italy treated with IL1 inhibition (IL1i).

Genomic analysis of 38 Legionella species identifies large and diverse effector repertoires

open access: yesNature Genetics, 2016
Infection by the human pathogen Legionella pneumophila relies on the translocation of ∼300 virulence proteins, termed effectors, which manipulate host cell processes. However, almost no information exists regarding effectors in other Legionella pathogens.

Internal Ubiquitin Electrophiles for Covalent Trapping and Inhibition of Deubiquitinases

open access: yesChemBioChem, EarlyView.
GA: Genetically encoded internal electrophiles in ubiquitin (Ub) enable the profiling of multivalent interaction interfaces, addressing a key limitation of current Ub‐based activity‐based probes. In this study, these probes capture a Ub–deubiquitinase interaction with minimal structural perturbation and serve as selective diUb probes that covalently ...

Whole genome sequence analysis reveals the broad distribution of the RtxA type 1 secretion system and four novel putative type 1 secretion systems throughout the Legionella genus. [PDF]

open access: yes, 2020
Type 1 secretion systems (T1SSs) are broadly distributed among bacteria and translocate effectors with diverse function across the bacterial cell membrane. Legionella pneumophila, the species most commonly associated with Legionellosis, encodes a T1SS at

Deubiquitination of phosphoribosyl-ubiquitin conjugates by phosphodiesterase-domain–containing Legionella effectors

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2019
Significance Ubiquitination is a vital posttranslational modification in eukaryotes. A variety of microbial pathogens exploit this pathway during their infection. Legionella pneumophila, the causative bacterial pathogen of Legionnaires’ disease, has been

Isolation of plasmids in Legionella pneumophila and Legionella-like organisms [PDF]

open access: yesInfection and Immunity, 1981
Agarose gel electrophoresis was employed to screen nine strains of Legionella-like bacteria and one strain of Legionella pneumophila for the presence of extrachromosomal deoxyribonucleic acid. Cryptic plasmids with molecular weights ranging from 46.6 x 10(6) to 59.8 x 10(6) were found in three of the isolates examined.

Development and Validation of a High‐Throughput Screening Assay for the Legionella ADP‐Ribosyl Transferase SdeA

open access: yesChemBioChem, EarlyView.
We show the optimization and application of a fluorogenic assay to monitor Legionella SdeA activity in a High‐Throughput‐Screening campaign. Ubiquitination of proteins is one of the most crucial post‐translational modifications in eukaryotic cells, typically involving conjugation of ubiquitin to a lysine residue in a substrate using a three‐enzyme ...

Presence of Legionellaceae in warm water supplies and typing of strains by polymerase chain reaction [PDF]

open access: yes, 2001
Outbreaks of Legionnaire's disease present a public health challenge especially because fatal outcomes still remain frequent. The aim of this study was to describe the abundance and epidemiology of Legionellaceae in the human-made environment.

How often is a work-up for Legionella pursued in patients with pneumonia? A retrospective study

open access: yesBMC Infectious Diseases, 2011
Background It is unclear how often patients with pneumonia are assessed for Legionella in endemic areas. Additionally, the sensitivity of the IDSA/ATS criteria for recommended Legionella testing is undefined.

Presence of Legionella spp. in cooling towers: the role of microbial diversity, Pseudomonas, and continuous chlorine application

open access: yesbioRxiv, 2019
Legionnaire’s Disease (LD) is a severe pneumonia caused by Legionella pneumophila. Cooling towers are the main source of L. pneumophila during large outbreaks. Colonization, survival, and proliferation of L.
