C2α‐carbanion‐protonating glutamate discloses tradeoffs between substrate accommodation and reaction rate in actinobacterial 2‐hydroxyacyl‐CoA lyase

open access: yesFEBS Open Bio, EarlyView.
Enzymes of the 2‐hydroxyacyl‐CoA lyase group catalyze the condensation of formyl‐CoA with aldehydes or ketones. Thus, by structural adaptation of active sites, practically any pharmaceutically and industrially important 2‐hydroxyacid could be biotechnologically synthesized. Combining crystal structure analysis, active site mutations and kinetic assays,

Effects of IGFBP4 deficiency on human preadipocyte proliferation and differentiation through the IGF1R/AKT pathway

open access: yesFEBS Open Bio, EarlyView.
IGFBP4 knockdown (KD) impairs preadipocyte proliferation and is associated with IGF1R protein downregulation and attenuated AKT phosphorylation. The mechanisms by which IGFBP4 KD influences the IGF1R/AKT signaling pathway involve newly synthesized proteins and lysosomal degradation pathways. Created in BioRender.

Optimized Ex Vivo Human Liver Slice Culture Maintains Extended Viability and Function for Hepatotoxicity Testing

open access: yesAdvanced Science, EarlyView.
An optimized ex vivo human liver slice culture system preserves tissue architecture, hepatocyte function, and immune‐stromal complexity for at least five days. By refining slice thickness, oxygenation, air‐liquid interface, and extracellular matrix support, the platform enables human‐relevant hepatotoxicity testing and mechanistic studies, offering a ...

COVID-19 pandemic and urban green spaces: Shifting usage behaviours and perceptions in Leipzig (Germany)?

open access: yesMoravian Geographical Reports
Crises such as the COVID-19 pandemic challenge some established human-landscape interactions notably. In this article, we analyse whether the pandemic had an impact on the perception of urban green spaces (UGS) and usage behaviours in Leipzig, Germany ...

MiR‐940 Suppresses Ferroptosis by Controlling Expression of Key Regulatory Genes

open access: yesAdvanced Science, EarlyView.
A CRISPR‐based screening identified miR‐940 as a critical suppressor of ferroptosis in cancer. By coordinating the downregulation of pro‐ferroptotic genes with the upregulation of GPX4, miR‐940 establishes a regulatory network that protects against ferroptosis and correlates with poor clinical outcomes in distinct cancer entities.

Adipocyte Myoglobin Is a Determinant of Energy Expenditure and a Potential Target to Limit Obesity

open access: yesAdvanced Science, EarlyView.
Myoglobin, known as a muscle oxygen‐carrying protein, is shown to play a key role in fat cells that burn energy. Loss of myoglobin reduces the body's ability to generate heat and increases obesity risk, while restoring it improves metabolism. The study identifies myoglobin as a regulator of fat burning and a potential target to enhance energy ...

“Importing” the Enlightenment and “Enlightenment at the margins”. The example of Bohemia as a language and religious frontier, 1770-1800

open access: yesCornova
This contribution explores the hypothesis that 18th-century Bohemia, which was effectively a political and informational periphery directed from Vienna, also functioned – thanks to its geographical position and other factors and despite unfavourable ...
