Genetic linkage analysis in the age of whole-genome sequencing
For many years, linkage analysis was the primary tool used for the genetic mapping of Mendelian and complex traits with familial aggregation. Linkage analysis was largely supplanted by the wide adoption of genome-wide association studies (GWASs). However,

Linkage analysis is used to map genetic loci using observations on relatives. It can be applied to both major gene disorders (parametric linkage) and complex diseases (model-free or non-parametric linkage), and it can be based on either a relatively small number of microsatellite markers or a denser map of single nucleotide polymorphisms (SNPs).

AbstractThis paper presents a methodology for the analysis of the linkage among the processes in an organization. The methodology has three main steps: model construction; connectivity analysis; and structure analysis. The model construction step generates a linkage matrix which is used in the following steps.

This paper introduces a simple method to characterize linkages among organizations in a system. Data required are collected by a questionnaire with scaled answers. This method can be applied to determine dominant and subordinate organizations, identify weak linkages among them, and give insights into linkage mechanisms to be established.

The mapping of human chromosomes, though much more complex, is related to cartography. To quote McKusick (1971), “We have a general picture of the various countries (the chromosomes) of the genetic world and have identified many of the municipalities (genes) on the basis of the specific traits to which they give rise.

SUMMARYThe estimation of the recombination fraction between pairs of loci on the X‐chromosome is discussed, with special reference to the bias from defined forms of ascertainment. Tables are presented which give lods corrected for the common forms of ascertainment. The interpretation of lods is discussed, and the concept of equivalent observations ...

Abstract Linkage analysis is the process of determining the approximate chromosomal location of a gene by looking for evidence of cosegregation with other genes whose locations are already known (i.e., marker genes). Cosegregation is a tendency for two or more genes to be inherited together, and hence for individuals with similar ...

Segregation and linkage analysis*
Animal Genetics, 1992Summary. Computer programs are available in the software package SAGE to perform a variety of segregation and linkage analyses used by human geneticists. These methods are designed specifically to uncover major gene segregation in pedigree data coming from non‐inbred populations.

Linkage analysis with inbreeding
Genetic Epidemiology, 1989AbstractFor computational reasons, linkage analysis of highly inbred pedigrees is often performed under the assumption that spouse pairs are genetically unrelated. A formulation of the likelihood of a pedigree which takes into account all of the genealogical information Ω, for an arbitrary relationship between a pair of spouses, is proposed. Lod scores

The triumph of linkage analysis
Annals of Neurology, 1990In the past decade, rapid advances have occurred in our understanding of the biochemical basis of inherited neurological disease, primarily resulting from detection of enzyme defects. In the next decade, the pulse of development in this area will quicken considerably because of the introduction of such recombinant DNA techniques as linkage analysis.
