ERK autoinhibition mechanism informs a drug combination strategy

open access: yesProtein Science, Volume 35, Issue 6, June 2026.
Abstract ERK is a key regulator in the MAPK pathway, controlling essential cell processes through dual‐phosphorylation‐based activation. We investigate the conformational equilibrium between inactive and active ERK states, which is controlled allosterically by its structurally unique C‐terminal L16 segment.

OpenPMX Software for Nonlinear Mixed‐Effect Models in Pharmacometrics: Precision Compared With NONMEM First‐Order Conditional Estimation

open access: yesCPT: Pharmacometrics &Systems Pharmacology, Volume 15, Issue 6, June 2026.
ABSTRACT Mixed effects models are a backbone of pharmacometrics, and NONMEM software, with the first‐order conditional method with interaction having become the de facto industry standard for model estimation. Documentation exists for the general mathematical methodology for estimation, but many technical and implementation details are lacking. OpenPMX

On Predicting Vulnerability Severity Using In‐Context Learning: An Industrial Case Study

open access: yesJournal of Software: Evolution and Process, Volume 38, Issue 6, June 2026.
ABSTRACT Modern software systems require earlier and more scalable vulnerability severity assessment to reduce exposure to high‐impact security flaws. Security analysts typically assign CVSS scores, but this manual triage does not scale with the growth of disclosed vulnerabilities and often depends on cloud LLM services that raise confidentiality ...

Finding Minimum‐Cost Explanations for Predictions Made by Tree Ensembles

open access: yesSoftware: Practice and Experience, Volume 56, Issue 6, Page 615-642, June 2026.
ABSTRACT The ability to reliably explain why a machine learning model arrives at a particular prediction is crucial when used as decision support by human operators of critical systems. The provided explanations must be provably correct, and preferably without redundant information, called minimal explanations.

Perspectives On Machine Learning Inference Serving in Real‐World Settings

open access: yesSoftware: Practice and Experience, Volume 56, Issue 6, Page 734-757, June 2026.
ABSTRACT Introduction As machine learning (ML)‐enabled systems become increasingly prevalent across industries, the engineering challenges of deploying and maintaining them in production have emerged as critical. The existing engineering knowledge base often derives from conceptual frameworks or case studies conducted by large technology companies ...
