How Do Loop Diuretics Act?

Drugs, 1991
In the thick ascending limb of the loop of Henle, NaCl reabsorption is mediated by a Na+/2Cl-/K+ cotransport system, present in the luminal membrane of this nephron segment. Loop diuretics such as furosemide (frusemide), piretanide, bumetanide and torasemide bind reversibly to this carrier protein, thus reducing or abolishing NaCl reabsorption.

Torsemide: A new loop diuretic

American Journal of Health-System Pharmacy, 1995
The pharmacology, pharmacokinetics, clinical efficacy, adverse effects, and dosage and administration of torsemide are reviewed. Torsemide belongs to the pyridine-sulfonylurea class of loop diuretics. Its primary site of activity is the thick ascending limb of the loop of Henle, where it blocks active reabsorption of sodium and chloride, resulting in ...

Ototoxicity of Loop Diuretics

Otolaryngologic Clinics of North America, 1993
The loop diuretics are drugs that increase the excretion of water and electrolytes in the urine by their action on the cells in the loop of Henle. Clinical reports of ototoxicity of these agents are reviewed, and the results of a number of studies in experimental animals are discussed.

Bumetanide, a new loop diuretic

Clinical Pharmacology & Therapeutics, 1976
The effect of bumetanide on renal function has been compared with that o.ffurosemide and a placebo in a double‐blind study in 9 healthy young men. The sequence for oral administration of the drug was subjected to a random assignation based upon the Latin‐square methodology under three different conditions.

Clinical Pharmacology of Loop Diuretics

Drugs, 1991
The clinical pharmacology of torasemide, bumetanide, piretanide and furosemide (frusemide) is discussed. These drugs share a similar mechanism of action in inhibiting Na(+)-K(+)-2Cl- reabsorption at the thick ascending limb of the loop of Henle. They differ in their routes of metabolism, pharmacokinetics, and potency.

Loop diuretics in heart failure

Heart Failure Reviews, 2011
Congestion is a major component of the clinical syndrome of heart failure, and diuretic therapy remains the cornerstone of congestion management. Despite being widely used, there is very limited evidence from prospective randomized studies to guide the prescription and titration of diuretics.
