eys+/-; lrp5+/− Zebrafish Reveals Lrp5 Can Be the Receptor of Retinol in the Visual Cycle [PDF]

open access: yesiScience, 2020
Summary: Vision is essential for vertebrates including humans. Sustained vision is accomplished by retinoid metabolism, the “visual cycle,” where all-trans retinol (atROL) is incorporated into the retinal pigment epithelium (RPE) from photoreceptors ...

Lrp5 functions in bone to regulate bone mass [PDF]

open access: greenNature Medicine, 2011
The human skeleton is affected by mutations in low-density lipoprotein receptor-related protein 5 (LRP5). To understand how LRP5 influences bone properties, we generated mice with osteocyte-specific expression of inducible Lrp5 mutations that cause high and low bone mass phenotypes in humans.

Lrp5 is not required for the proliferative response of osteoblasts to strain but regulates proliferation and apoptosis in a cell autonomous manner. [PDF]

open access: yesPLoS ONE, 2012
Although Lrp5 is known to be an important contributor to the mechanisms regulating bone mass, its precise role remains unclear. The aim of this study was to establish whether mutations in Lrp5 are associated with differences in the growth and/or ...

Zebrafish mutants reveal unexpected role of Lrp5 in osteoclast regulation

open access: yesFrontiers in Endocrinology, 2022
Low-density Lipoprotein Receptor-related Protein 5 (LRP5) functions as a co-receptor for Wnt ligands, controlling expression of genes involved in osteogenesis.

TIEG1 is upregulated in Lrp5/6‐mediated valve osteogenesis [PDF]

open access: greenJournal of Cellular Biochemistry, 2018
AbstractWe have previously demonstrated that Lrp5/6/β‐catenin plays an important role in valve calcification with a specific osteogenic phenotype defined by increased bone mineral content and overall valve thickening. Recent studies indicate that TIEG1 may be involved in mediating the Wnt signaling pathway in bone, which is known to play critical roles

The LRP5 High-Bone-Mass G171V Mutation Disrupts LRP5 Interaction with Mesd [PDF]

open access: yesMolecular and Cellular Biology, 2004
The mechanism by which the high-bone-mass (HBM) mutation (G171V) of the Wnt coreceptor LRP5 regulates canonical Wnt signaling was investigated. The mutation was previously shown to reduce DKK1-mediated antagonism, suggesting that the first YWTD repeat domain where G171 is located may be responsible for DKK-mediated antagonism.

LRP5 variants may contribute to ADPKD [PDF]

open access: yesEuropean Journal of Human Genetics, 2015
Mutations in Polycystic Kidney Disease proteins (PKD1 or PKD2) are causative for autosomal dominant polycystic kidney disease (ADPKD). However, a small subset of ADPKD probands do not harbor a mutation in any of the known genes. Low density lipoprotein Receptor-related Protein 5 (LRP5) was recently associated with hepatic cystogenesis in isolated ...

LRP5 gene polymorphism and cortical bone [PDF]

open access: yesAging Clinical and Experimental Research, 2010
There is evidence that distinct genetic polymorphisms of LRP5 are associated with low Bone Mineral Density (BMD) and the risk of fracture. However, relationships between LRP5 polymorphisms and micro- and macro architectural bone characteristics assessed by pQCT have not been studied.
