Optogenetic‐Controlled iPSC‐Based Vaccines for Prophylactic and Therapeutic Tumor Suppression in Mice

open access: yesAdvanced Science, EarlyView.
This study develops an a red/far‐red light‐controlled iPSC‐based vaccines (RIVA) based on the chimeric photosensory protein FnBphP and its interaction partner LDB3 for cancer immunotherapy. The RIVA preserves the multiple intrinsic tumor antigens of iPSCs and enables optogenetic control of the expression of the immune adjuvant type I interferon β under

Single‐Cell RNA Sequencing Reveals the Heterogeneity in Differentiation Trajectory and Tumor Microenvironment Leading to More Aggressive Phenotypes of Papillary Thyroid Cancer in Children and Young Adult Patients

open access: yesAdvanced Science, EarlyView.
scRNA sequencing reveals distinct tumor ecosystem features in CAYA‐PTC, marked by enriched CD4T_Tfh, CD8T_Tex cells, and rapid malignant thyrocyte differentiation (absent of mild‐state BRAF‐like population) toward invasive states. Prominent emCAF_LAMP5 populations interact with endothelial cells and thyrocytes facilitate angiogenesis and metastasis ...

RELA Ablation Contributes to Progression of Hepatocellular Carcinoma with TP53R249S Mutation and is a Potential Therapeutic Target

open access: yesAdvanced Science, EarlyView.
Genome‐wide CRISPR/Cas9 based screening identified RELA as a key tumor suppressor in TP53R249S‐mutant HCC. Its loss promotes tumorigenesis and metastasis via DVL1‐mediated Wnt/β‐catenin activation, while its agonist betulinic acid suppresses tumor progression.

Role of inducible nitric oxide (iNOS) and nitrosative stress in regulating sex differences in secondary lymphedema

open access: yesFrontiers in Physiology
Secondary lymphedema is a common complication following surgical treatment of solid tumors. Although more prevalent in women due to higher breast cancer rates, men also develop lymphedema, often with more severe manifestations.
