Real World Lab Data: Patterns of Lymphocyte Counts in Fingolimod Treated Patients
Objective:Fingolimod is approved for the treatment of highly active relapsing remitting multiple sclerosis (MS) patients and acts by its unique mechanism of action via sphingosine-1-phosphate receptor-modulation.

Concepts of GPCR-controlled navigation in the immune system
G-protein-coupled receptor (GPCR) signaling is essential for the spatiotemporal control of leukocyte dynamics during immune responses. For efficient navigation through mammalian tissues, most leukocyte types express more than one GPCR on their surface ...

Th17 responses are not altered by natural exposure to seasonal allergens in pollen-sensitive patients [PDF]
Background: Allergic rhinitis affects 10–30 % of the global population and this number is likely to increase in the forthcoming years. Moreover, it commonly co-exists with allergic asthma as a chronic allergic respiratory syndrome. While the involvement

DCAF13 Safeguards Hematopoietic Stem Cells via RRS1‐Regulated Ribosome Biogenesis
This study establishes DCAF13 as an essential regulator for hematopoietic stem cell (HSC) function. Its deletion in mice causes lethal pancytopenia and HSC depletion. Mechanistically, DCAF13 interacts with RRS1 and mediates its non‐degradative K27‐linked ubiquitination, thereby stabilizing RRS1 to maintain ribosome biogenesis and protein translation ...

Improved Systemic Immunochemotherapy Employing an Oxaliplatin‐TLR7/8 Agonist Prodrug Strategy
A platinum(IV) prodrug is developed to systemically deliver the TLR7/8 agonist gardiquimod. Tumor‐targeting is mediated by an albumin‐binding maleimide, which limits premature complex activation. Ox‐Gardi‐Mal accumulates specifically in the tumor, where it activates the immune system.

Persistent attenuation of lymphocyte subsets after mass SARS-CoV-2 infection
Objectives: Growing evidence suggests that lymphocyte subsets are declined in COVID-19 patients, but it is unclear if these alterations persist after widespread exposure to SARS-CoV-2 or how long they last.

Multi-parameter immune profiling of peripheral blood mononuclear cells by multiplexed single-cell mass cytometry in patients with early multiple sclerosis [PDF]
Multiple sclerosis (MS) is an inflammatory demyelinating and neurodegenerative disease of the central nervous system (CNS). Studies in rodent models demonstrated an association of CNS-infiltrating monocyte-derived macrophages with disease severity ...

This study reveals that m6A regulators cooperatively upregulate BGN in melanoma, promoting malignancy. Within the tumor microenvironment, CAFs show highest BGN expression. The BGN/MDK axis mediates cancer‐stroma crosstalk, driving normal fibroblast (NF) activation and enhancing the pro‐tumor effect of CAFs, highlighting a promising therapeutic target ...

Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.
