Mechanically Regulated Secretion: How Physical Forces Instruct the Secretory Pathway and Remodel the Secretome

open access: yesAdvanced Science, EarlyView.
Mechanical cues, including ECM stiffness, stretch, compression, shear stress, and traction forces, remodel trafficking across the secretory pathway, from the ER and Golgi to endolysosomal compartments and the plasma membrane. The resulting secretome reshapes ECM composition and tissue mechanics, establishing feedback loops that support homeostasis or ...

Stabilizing Functional Nucleic Acids for Real‐Sample Diagnostics

open access: yesAngewandte Chemie, EarlyView.
Functional nucleic acids enable programmable real‐sample diagnostics, but often lose function in complex biological matrices. This review highlights four stabilization levers—chemical, topological, architectural, and microenvironment/interface engineering—that preserve aptamer and DNAzyme performance in saliva, serum, urine, and stool, reframing ...

Bacteria‐Derived 1‐Allylpiperazine Alleviates Symptoms of Parkinson's Disease Model by Promoting Autophagy

open access: yesAdvanced Science, EarlyView.
Parkinson's disease currently has no disease‐modifying therapies. In Caenorhabditis elegans, Escherichia coli E8 alleviates α‐synuclein‐provoked PD‐related defects through its bioactive metabolite 1‐ALPP, which combats PD by triggering AMPK/mTOR/HLH‐30‐dependent autophagy and suppressing α‐synuclein LLPS‐mediated fibrillization.

β‐Adducin Restrains PLA2G4A‐Dependent Lysosomal Membrane Permeabilization and Neuronal Death in Ischemic Stroke

open access: yesAdvanced Science, EarlyView.
This study identifies β‐adducin as a regulator of lysosomal integrity in ischemic neurons. Ischemia‐associated FTO loss increases m6A modification and destabilizes Add2 mRNA, reducing β‐adducin expression and promoting PLA2G4A accumulation at lysosomes.

Chlorination‐Driven BODIPY Fluorescent Probes for the Selective Monitoring of Myeloperoxidase Activity in Cells and Inflamed Mouse Models

open access: yesAngewandte Chemie, EarlyView.
A meso‐carboxamide‐substituted BODIPY fluorescent probe reports myeloperoxidase (MPO) activity via electrophilic chlorination by MPO‐derived HOCl, generating a highly emissive chlorinated product with a ca. 40 nm bathochromic shift. Applications in quantitative MPO assays, cellular imaging of MPO‐derived HOCl, redox‐dependent discrimination of cancer ...

Low‐Density Lipoprotein Receptor‐Targeted Biomimetic Nanoplatform Suppresses Glioblastoma Stemness and Remodels Antitumor Immunity via Dual Metabolic Disruption

open access: yesAdvanced Science, EarlyView.
A low‐density lipoprotein receptor‐oriented biomimetic nanoplatform couples brain‐tumor delivery with a mechanistically matched regorafenib‐erythrodiol combination. Concurrent disruption of autophagic nutrient salvage and cholesterol homeostasis collapses bioenergetic and signaling support, suppresses glioblastoma stemness, and promotes immunogenic ...

Age‐Related Lamin B1 Deficiency Activates SLC7A11‐Dependent Disulfidptosis to Impair Osteogenesis via Intercepting Mitochondrial Fission

open access: yesAdvanced Science, EarlyView.
This study reveals that age‐related LMNB1 deficiency in mesenchymal stem cells epigenetically upregulates SLC7A11, triggering actin disulfidptosis and mitochondrial dysfunction. This cascade impairs bone formation in senile osteoporosis. Furthermore, researchers identified the FDA‐approved drug naldemedine as a targeted SLC7A11 inhibitor to reverse ...

Differential Diagnosis of Mucopolysaccharidoses Types I, II, III, IVA, and VI Through Analysis of Leukocytic Inclusions

open access: yesAmerican Journal of Medical Genetics Part A, EarlyView.
ABSTRACT Mucopolysaccharidoses (MPS) are lysosomal storage disorders characterized by the accumulation of glycosaminoglycans (GAGs), which can lead to cytoplasmic alterations in leukocytes. The objective of this study was to characterize leukocyte inclusions in patients with different types of MPS and assess their diagnostic relevance.
