This chapter focuses on a method of sequencing M13 clones that allows the determination of the nucleotide sequence of inserts cloned into M13 vectors. A 17-mer oligonucleotide primer is annealed to the M13 vector in a position adjacent to the polylinker cloning region.

Replication of the Small Coliphage M13: Evidence for Long-living M13 Specific Messenger RNA
Nature, 1970THE stability of messenger RNA has been the object of intense investigations. In the Escherichia coli system, predominantly the mRNAs for inducible enzymes or mRNAs specified by lytic or inducible phages have been studied. All such mRNAs were found to have half lives of the order of several minutes, even when measured on the basis of physical rather ...

Growing Bacteriophage M13 in Liquid Culture
Cold Spring Harbor Protocols, 2006Stocks of bacteriophage M13 are usually grown in liquid culture. The infected bacteria do not lyse but, instead, grow at a slower than normal rate to form a dilute suspension. The inoculum of bacteriophage is almost always a freshly picked plaque or a suspension of bacteriophage particles obtained from a single plaque, as described here. Infected cells

The initial steps in infection with coliphage M13
Virology, 1964Abstract Infection of Escherichia coli with the rod-shaped DNA bacteriophage M13 begins with an irreversible attachment of the phage to one of a very limited number of sites on the cell surface. The attachment rate is maximal in media of molar ionic strength of about 0.1.
