Liquid biopsy‐based diagnostic evaluation of hypermethylated CpG sites for ovarian cancer diagnosis

open access: yesMolecular Oncology, EarlyView.
This schematic outlines the workflow from biomarker identification to duplex MethyLight assay validation for epithelial ovarian cancer diagnosis using cfDNA‐based liquid biopsy. Initial screening of hypermethylated CpG candidates (cg02957270, cg10061138 cg00480298, COL2A1) was performed in tissue using ARMS‐PCR, COBRA, qPCR and image analysis. Selected

Analysis of the Structures of Mating-Type A and B Loci in Stropharia rugosoannulata Based on Genomic Data and Development of SNP Molecular Markers

open access: yesHorticulturae
Stropharia rugosoannulata is a widely cultivated edible fungus with high economic and nutritional value. It is a tetrapolar heterothallic basidiomycete. The development of single nucleotide polymorphism (SNP) markers for mating-type identification holds ...

Rapid and Efficient CRISPR/Cas9-Based Mating-Type Switching of Saccharomyces cerevisiae

open access: yesG3: Genes, Genomes, Genetics, 2018
Rapid and highly efficient mating-type switching of Saccharomyces cerevisiae enables a wide variety of genetic manipulations, such as the construction of strains, for instance, isogenic haploid pairs of both mating-types, diploids and polyploids. We used

A Third Multiallelic Mating-type Locus in Physarum polycephalum

open access: yesMicrobiology, 1987
SUMMARY: Sexual development (crossing) in Physarum polycephalum occurs when two haploid amoebae fuse to form a diploid plasmodium. The imz locus influences the maximum pH at which crossing can occur. A new allele of imz has been identified, bringing the total number of alleles to three.

Epigenetic heterogeneity and plasticity in therapy‐induced tumor states through single‐cell multi‐omics

open access: yesMolecular Oncology, EarlyView.
Single‐cell multi‐omics reveals epigenetic heterogeneity across therapy‐adaptive tumor states, including quiescent/dormant, drug‐tolerant persister, and EMT‐like phenotypes. By linking regulatory features with state‐associated biomarkers, these approaches inform biomarker‐guided therapeutic strategies for evolving tumors.

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Genotyping of the MTL loci and susceptibility to two antifungal agents of Candida glabrata clinical isolates

open access: yesMemorias do Instituto Oswaldo Cruz, 2009
The opportunistic fungal pathogen Candida glabrata is the second most common isolate from bloodstream infections worldwide and is naturally less susceptible to the antifungal drug fluconazole than other Candida species. C.

Yeast mismatch repair components are required for stable inheritance of gene silencing.

open access: yesPLoS Genetics, 2020
Alterations in epigenetic silencing have been associated with ageing and tumour formation. Although substantial efforts have been made towards understanding the mechanisms of gene silencing, novel regulators in this process remain to be identified.

Epigenetic silencing of the liver‐specific lncRNA LUNAR promotes liver cancer progression via NOTCH activation

open access: yesMolecular Oncology, EarlyView.
LUNAR is a liver‐specific long noncoding RNA (lncRNA) that is highly expressed in normal liver but becomes epigenetically silenced in hepatocellular carcinoma through promoter hypermethylation. Loss of LUNAR is associated with NOTCH activation, epithelial–mesenchymal transition, and metastasis, whereas restoring LUNAR restrains metastatic progression ...

Comparative Analysis of Mating Behavior Characteristics of Mutants at the white Locus of Drosophila melanogaster [PDF]

open access: yesУчёные записки Казанского университета: Серия Естественные науки, 2017
Drosophila melanogaster serves as an excellent model system to study the control of innate behaviors. Behavior is the ability of animals to change their actions under the influence of internal and external factors, a characteristic feature of the animal ...
