6日間暗所発芽させた黒緑豆種子よりCPaseを精製し, その酵素化学的性質を検討した。1) 粗酵素液を硫安分画 (30~70%), Sephadex G-150ゲルろ過, DEAE-Sepharoseクロマトグラフィー, SephadexG-150再ゲルろ過, DBAE-Sepharose再クロマトグラフィーおよびTOYOPEARL HW-50Fゲルろ過を行なうことにより, ディスク電気泳動的に均一なCP-ase標品を単離した。収率は7.8%であり比活性の上昇率は110倍であった。ゲルろ過法による分子量は約7万と推定された。2) 精製CPaseのZ-Phe-Ala水解時における至適pHは6.0, 至適温度は50℃であった。室温2時間放置時の安定pH領域は5~8であり, 10分間熱処理時の耐熱性は50℃
