Purification and Some Properties of Carboxypeptidase from Black Matpe Seedlings
Eiyo To Shokuryo, 1982 6日間暗所発芽させた黒緑豆種子よりCPaseを精製し, その酵素化学的性質を検討した。1) 粗酵素液を硫安分画 (30～70%), Sephadex G-150ゲルろ過, DEAE-Sepharoseクロマトグラフィー, SephadexG-150再ゲルろ過, DBAE-Sepharose再クロマトグラフィーおよびTOYOPEARL HW-50Fゲルろ過を行なうことにより, ディスク電気泳動的に均一なCP-ase標品を単離した。収率は7.8%であり比活性の上昇率は110倍であった。ゲルろ過法による分子量は約7万と推定された。2) 精製CPaseのZ-Phe-Ala水解時における至適pHは6.0, 至適温度は50℃であった。室温2時間放置時の安定pH領域は5～8であり, 10分間熱処理時の耐熱性は50℃

Changes of Proteins and Enzyme Activity in Black Matpe Organs during Germination.
Nippon Eiyo Shokuryo Gakkaishi, 1997 ブラックマッペの発芽にともなう各部位のタンパク質の変化を, ゲル濾過法を用い検討した。同時にプロテアーゼインヒビター活性およびプロテアーゼ活性を調べ, タンパク質の変化と酵素活性の変動との関連性をも検討した。さらに7日発芽体の部位別の, 遊離アミノ酸組成を調べたところ以下のことが明らかとなった。1) 発芽にともない, 子葉のタンパク質量は減少したが, 胚軸では, 発芽とともにわずかではあるが増加した。2) 未発芽豆の子葉と胚芽のタンパク質組成を比較したところ, 三つのピークが認められ, 子葉においてその比率は, 分子量の大きいものから順に, 7: 2: 1, 胚芽では4: 5: 1であった。また貯蔵タンパク質と思われるピーク1 (分子量の最も大きいもの) について ...
