Menin Inhibitor Induced Menin Protein Degradation Contributes to Menin Inhibitor Efficacy

Blood, 2023
The chromatin adaptor protein Menin ( MEN1) is necessary for sustained proliferation in acute leukemias driven either by Lysine Methyltransferase 2A (MLL1/ KMT2A) gene rearrangements. Small molecule Menin inhibitors (MIs) disrupt the MENIN-KMT2A protein interaction by competing with KMT2A for a binding pocket in Menin.

Menin and its interacting proteins: elucidation of menin function

Trends in Endocrinology & Metabolism, 2006
The multiple endocrine neoplasia type 1 (MEN1) gene is a tumor suppressor gene encoding a 610 amino acid nuclear protein, menin. Although mutations of the MEN1 gene are responsible for MEN 1 syndrome, the intracellular functions of menin have not been fully elucidated.

The Menin Gene

2009
Multiple endocrine neoplasia type I (MEN-1) is an autosomal dominant syndrome featuring tumors of endocrine origin. Heterozygous germline mutations in the MEN-1 tumor suppressor gene predispose MEN-1 patients to tumor development, mainly in parathyroid, pancreatic islet cells, and the anterior pituitary gland. Since the MEN-1-encoded protein, menin, is

Cellular Functions of Menin

2009
Since its discovery as a novel protein some 10 years ago, many cellular functions of menin have been identified. However, which ones of these relate specifically to menin's role as a tumor suppressor and which ones not remains unclear. Menin is predominantly nuclear and acts as a scaffold protein to regulate gene transcription by coordinating chromatin

Menin and bone metabolism

Journal of Bone and Mineral Metabolism, 2012
Menin, a product of the MEN1 gene, is related to the ontogeny of several cancers such as MEN1 and sporadic endocrine tumors, although it is considered to be a tumor suppressor. Many proteins interact with menin, and it is involved in various biological functions in several tissues.

Role of Menin in Bone Development

2009
Menin function is related to transcriptional regulation and cell cycle control and it physically and functionally interacts with osteotropic transcription factors, such as Smad1/5, Smad3, Runx2 and JunD. Menin promotes the commitment of pluripotent mesenchymal stem cells to the osteoblast lineage, mediated by interactions between menin and the BMP ...

Menin: from molecular insights to clinical impact

open access: yesEpigenomics
Menin, the protein product of the MEN1 gene, is essential for development and has been implicated in multiple different cancer types. These include leukemias and several different solid tumors, including neuroendocrine tumors.

At the Menin Gate

Journal of Historical Sociology, 1996
Abstract This essay reports an historical and sociological journey through family memories to the battlefield of World War I. It follows the short military career of a working class lad through the Territorial Army (Northumberland Fusiliers) to his death on the Ypres Salient in 1915.

Role of Menin in Neuroendocrine Tumorigenesis

2009
The menin protein encoded by the MEN1 tumor suppressor gene is ubiquitously expressed and highly conserved evolutionarily. The combination of findings from current in vitro and in vivo studies has not yielded a comprehensive understanding of the mechanisms of menin's tumor suppressor activity or the specific role for menin in endocrine tumorigenesis ...
