A dnaK homolog in the archaebacterium Methanosarcina mazei S6

A fragment of genomic DNA cloned from the methanogenic archaebacterium, Methanosarcina mazei strain S6, was found to contain an 1857-bp open reading frame (ORF). A sequence matching the consensus ribosome-binding sequence determined for other methanogens was found upstream from the ORF.

2-(Methylthio)ethanesulfonate (CH3S-CoM) is formed as an intermediate in methanogenesis from methanol by cell-free extracts of Methanosarcina barkeri. The enzyme system involved in the methyl transfer from methanol to 2-mercaptoethanesulfonate (HS-CoM) was resolved into two enzyme fractions.

AbstractBiologically‐produced CH4 derives from either the reduction of CO2 or the methyl group of acetate by two separate pathways present in anaerobic microbes from the Archaea domain. Elucidation of the pathway for CO2 reduction to CH4, the first to be investigated, has yielded several novel enzymes and cofactors.
