Cytostatic active methylhydrazine derivatives split off formaldehyde after mild dehydrogenation with potassium hexacyanoferrate (III). It is suggested that the primary formedazomethines andN-hydroxymethylderivatives participate in cytostatic efficacy by their alkylating ability (aminomethylation).

Initial experiences with methylhydrazine, a new cytotoxic agent

The authors present their results using methylhydrazine (‘Ro 4-6467’, ‘Natulan’) in the treatment of 40 patients with malignant disease. The progress of 29 patients could be adequately assessed. Remissions occurred frequently in patients with Hodgkin's disease and some solid tumours also showed a response.

SummaryResults of Chemotherapic treatment in Hodgkin's disease are reported. 23 cases were treated by Vincaleucoblastine, 19 by Methylhydrazine, and 12 by both drugs. With VLB we obtained 6 complete remissions, 12 partial remissions and 5 failures; with MH the remissions were 9 complete, 4 partial and 6 failures.

Reaction of methylhydrazine with dimethyl chlorothiophosphate

The reaction of 2 equivalents of methylhydrazine with 1 equivalent of (MeO)2P(S)Cl at 0 °C produced the phosphorus hydrazide (MeO)2P(S)NMeNH21 in good yield. However, at 25 °C a mixture of three products was obtained according to 1H and 31P NMR spectra. The pure individual components were separated by column chromatography.
