Stoichiometric Binding of Cyclophilin‐A to The HIV‐1 Capsid Modulates Its Mechanoelastic Properties

open access: yesSmall, EarlyView.
The HIV‐1 capsid is a protein nanocapsule that must balance rigidity and flexibility to enter the cell nucleus. Atomic force microscopy simulations reveal that binding of the host protein Cyclophilin A tunes this mechanical response: partial coverage preserves deformability, while excessive binding induces brittleness and failure.

Chemical and Biological Determinants of PEG‐Lipid Shedding From Therapeutic Nanoparticles

open access: yesSmall, EarlyView.
PEG‐lipids play a central role in governing lipid nanoparticle stability, targeting, and function. However, PEG‐lipids can “shed” from the nanoparticle surface in biological environments, with the extent and kinetics of shedding influenced by chemical and biological factors.

Rapid and Efficient Affinity Isolation of Engineered Extracellular Vesicles Encapsulating Functional Proteins via Internal Twin‐Strep Tetraspanin Tagging

open access: yesSmall, EarlyView.
Structure‐guided internal twin‐Strep tagging of the tetraspanins CD63 or CD9 enables rapid and efficient antibody‐free affinity isolation of engineered extracellular vesicles (EVs) using Strep technology while allowing luminal protein loading. The approach removes over 96% of contaminants, separates tagged EVs from coexisting viral particles, and ...

Cardiomyocyte Membrane‐Camouflaged Spinach Nano‐Thylakoids Enhance Myocardial Energy Metabolism and Ameliorate Heart Failure

open access: yesSmall, EarlyView.
Schematic illustration of the construction of MM‐NTU, its cardiac‐targeting behavior, and its light‐activated therapeutic mechanism. Thylakoids isolated from spinach chloroplasts were processed into nano‐thylakoid units (NTU) and further camouflaged with cardiomyocyte membranes to generate MM‐NTU.

Proximity Proteomics Maps Candidate Cellular Uptake Pathways for Cell‐Derived Nanovesicles

open access: yesSmall Methods, EarlyView.
TurboID‐based dual‐sided proteomics compares mechanically generated cell‐derived nanovesicles with naturally secreted small extracellular vesicles. Vesicle proteomes define candidate interaction signatures, while recipient‐cell proximity labelling identifies exposure‐associated proteins.
