Structure and stereochemistry of the base excision repair glycosylase MutY reveal a mechanism similar to retaining glycosidases. [PDF]
MutY adenine glycosylases prevent DNA mutations by excising adenine from promutagenic 8-oxo-7,8-dihydroguanine (OG):A mismatches. Here, we describe structural features of the MutY active site bound to an azaribose transition state analog which indicate a

Dissecting conformational contributions to glycosidase catalysis and inhibition [PDF]
Glycoside hydrolases (GHs) are classified into >100 sequence-based families. These enzymes process a wide variety of complex carbohydrates with varying stereochemistry at the anomeric and other ring positions.

GASdb: a large-scale and comparative exploration database of glycosyl hydrolysis systems [PDF]
Background The genomes of numerous cellulolytic organisms have been recently sequenced or in the pipeline of being sequenced. Analyses of these genomes as well as the recently sequenced metagenomes in a systematic manner could possibly lead to ...

The tetrasaccharide linkage region of proteoglycans is synthesized through a series of well‐controlled enzymatic reactions in nature. The expression, activity, and substrate profile of these biosynthetic enzymes and the latest development of chemoenzymatic synthesis of linkage region‐bearing glycopeptides are summarized in this review.

Stereochemical course of the hydrolysis reaction catalyzed by chitinases Al and D from Bacillus circulans WL-12 [PDF]
Chitinases A1 and D were purified from the periplasmic proteins produced by Escherichia coli HB101 harbouring recombinant plasmids carrying respectively the chiA and chiD genes of Bacillus circulans WL-12.

A double photochemical cryo thiol‐ene coupling process was developed as a general method for the synthesis of 1,2‐cis‐α,α’‐thio‐linked di‐ and oligosaccharides. The procedure includes i) UV‐initiated coupling reaction of 2‐substituted glycals with HSAc in AcOH at −80 °C in frozen state, ii) selective S‐deacetylation of the resulting glycosyl‐α‐SAc ...

The regions surrounding the catalytic amino acids previously identified in a few "retaining" O-glycosyl hydrolases (EC 3.2.1) have been analyzed by hydrophobic cluster analysis and have been used to define sequence motifs. These motifs have been found in

New α‐glycosides of chalcones and dihydrochalcones, analogues of naturally occurring phlorizin, have been synthesized using a CGTase‐catalyzed glycosylation that was successfully scaled‐up in a 10‐liter reactor. The α‐glycosides were (i) purified using an original centrifugal partition chromatography technique, (ii) characterized through NMR and LC–MS ...
