Repurposing Cardiac Glycosides to Potentiate CD47 Blockade through Calreticulin‐mediated Phagocytic Effects for Lung Cancer Treatment

Repurposing cardiac glycosides (CGs) significantly strengthens the efficacy of anti‐CD47 antibody for lung cancer treatment by boosting macrophage phagocytosis. This novel combination blocks the “don't eat me” signal and increases dual “eat‐me” signals, Fc‐FcγR and cell surface expression of calreticulin (ecto‐CRT).

Physiological flow (0.5 Pa) conditioning promotes resilience of an iPSC‐derived blood–brain barrier endothelium to mild systemic inflammation. In contrast, high “cytokine storm” levels induce barrier breakdown, fibrinogen leakage, and astrocyte activation, reconstituting the two‐hit injury mechanism on‐a‐chip.

cGAS Inhibits ALDH2 to Suppress Lipid Droplet Function and Regulate MASLD Progression

cGAS is a cytosolic DNA sensor known for innate immune defense. cGAS binds and suppresses ALDH2 are showed, limiting acetyl‐CoA production. cGAS loss activates ALDH2, promoting histone acetylation, lipogenesis, and lipid droplets accumulation. cGas‐/‐ mice on a high‐fat diet show signs of worsened MASLD.
