ABSTRACT Chronic rhinosinusitis with nasal polyps (CRSwNP) exhibits pronounced endotypic heterogeneity, with macrophages serving as key drivers of sustained mucosal inflammation. In this study, we identify S100A9 as a macrophage‐derived alarmin that is markedly elevated in CRSwNP tissues.

“Super‐Responders” to Dupilumab Treatment in Patients With Primary Diffuse Chronic Rhinosinusitis With Nasal Polyps

This study aimed to evaluate the feasibility of extending dupilumab dosing intervals in T2‐CRSwNP by assessing how many patients maintain disease control with once‐every‐12‐weeks (Q12W) dosing and comparing clinical characteristics of super‐responders and excellent responders. Tapering of dupilumab to Q12W appears feasible in super‐responding patients.

IL‐33 Drives Inflammatory Changes and Extracellular Trap Formation in Eosinophils Involving Oxidised LDL and Complement Pathways

IL‐33 induces inflammatory gene signatures in eosinophils, characterised by the upregulation of cell surface markers, inflammatory mediators and regulatory factors, all of which were also observed in nasal polyp‐derived eosinophils. Effect of Dupilumab on Airway Inflammation in Patients With Persistent Asthma

Efficacy and Safety of Dupilumab in Chinese Adult Patients With Chronic Rhinosinusitis With Nasal Polyps: A Randomized, Placebo‐Controlled, Phase III Trial

Dupilumab significantly improved nasal polyp score, nasal congestion, olfaction, total symptom score, and disease‐specific quality of life in Chinese patients with uncontrolled CRSwNP. Incidence of treatment‐emergent adverse events was similar between dupilumab and placebo.
