Abstract Reaction of Mes NPC1 with triphenylcarbenium salts (BF4 or PF6) produces a difluorophosphine and not the expected iminophosphenium cation. This compound then undergoes an Arbusov-type rearrangement to generate a difluoro-iminophosphorane.

Niemann Pick C (NPC) is a fatal hereditary neurovisceral disorder associated with a progressive loss of neurons of unknown mechanism. The disease is caused by mutation in either of two genes, termed npc1 and npc2, accounting for ∼95% and ∼5% of patients, respectively. Recent data suggest a cell-autonomous cause for neuronal cell death.

NPC1‐mediated cholesterol export from lysosomes

Endocytosed, LDL‐derived cholesterol is exported from the interior of lysosomes by a process that requires the action of NPC1 and NPC2 proteins. We have recently shown that LAMP1 and LAMP2 proteins interact with NPC1 and NPC2 and also bind cholesterol (1).
