This study identifies ARID1B as a chromatin‐bound driver of tumor growth in TNBC. ARID1B impairs ARID1A function and directly activates oncogenic programs through SWI/SNF remodeling. Its nuclear localization, mediated by the KPNA2–KPNB1–RANBP2 import machinery, is essential for its tumor‐promoting activity.

The VRK1 protein kinase regulates the phosphorylation of BAF, which binds to dsDNA and LEM domain–containing proteins. VRK1 depletion increases the immobile fraction of BAF at the nuclear periphery and disturbs nuclear envelope architecture.

In this study, a novel mechanism is unveiled by which ADD1, acting as a molecular switch, coordinates actin branch dynamics and the transport of endocytic viruses and cargoes. Phosphorylation of ADD1 at Ser726 reduces actin branch density, enhancing endosome fusion and attachment to microtubules.

This study utilizes patient‐derived osteosarcoma organoids and CRISPR screening to identify ERCC6 and HNRNPM as key mediators of cisplatin resistance via the PI3K/AKT pathway and BAX alternative splicing.
