Nucleosome Mapping

2009
The basic repeating unit of chromatin, the nucleosome, is known to play a critical role in regulating the process of gene transcription. The positioning of nucleosomes on a promoter is a significant determinant in its responsiveness to gene-inducing signals.

Dynamic nucleosomes

Chromosome Research, 2006
It is now widely recognized that the packaging of genomic DNA together with core histones, linker histones, and other functional proteins into chromatin profoundly influences nuclear processes such as transcription, replication, repair and recombination. How chromatin structure modulates the expression and maintenance of knowledge encoded in eukaryotic

Nucleosome Positioning Determinants

Journal of Molecular Biology, 2007
A previous report demonstrated that one site in a nucleosome assembled onto a synthetic positioning sequence known as Fragment 67 is hypersensitive to permanganate. The site is required for positioning activity and is located 1.5 turns from the dyad, which is a region of high DNA curvature in the nucleosome.

Remodeling of Individual Nucleosomes in Nucleosome Arrays

Adenosin triphosphate (ATP)-dependent nucleosome remodeling factors sculpt the nucleosomal landscape of eukaryotic chromatin. They deposit, evict, or reposition nucleosomes along DNA in a process termed nucleosome sliding. Remodeling has traditionally been analyzed using mononucleosomes as a model substrate.

Apoptosis and nucleosomes

Lupus, 2002
Drs Berden and Koutouzov presented evidence that nucleosomes are antigens in lupus pathogenesis and that apoptotic cells are the source of nucleosomes. Berden's group measured persistence of circulating nucleosomes and nucleosome–antibody complexes in autoimmune mice and demonstrated nucleosomaldepositionin skin of SLE patientsas well as in renal ...

Packaging without nucleosomes

Science, 2017
Chromosomes The fundamental compaction of DNA in chromosomes is thought to start with the wrapping of DNA around nucleosomes, but is nucleosome wrapping required for the condensation of mitotic chromosomes? Shintomi et al. combined Xenopus egg extracts and mouse sperm nuclei and found that chromosome-like structures could be assembled in the near ...
