Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

Numerical Integrators That Preserve Symmetries and Reversing Symmetries

open access: yes, 1996
We consider properties of flows, the relationships between them, and whether numerical integrators can be made to preserve these properties. This is done in the context of automorphisms and antiautomorphisms of a certain group generated by maps ...

Properties of Hamiltonian Variational Integrators [PDF]

open access: yes, 2017
This dissertation explores Hamiltonian variational integrators. Variational integrators are a common class of symplectic integrators, which have primarily been analyzed and constructed by discretizing Hamilton's principle.

Stimulator of interferon genes agonist augmented antitumor immunity of osimertinib in Egfr‐mutated lung cancer

open access: yesMolecular Oncology, EarlyView.
Combining osimertinib with the STING agonist ADU‐S100 activates innate and adaptive immunity to overcome the non‐inflamed microenvironment of Egfr‐mutant lung cancer. This combination increases NK and CD8+ T‐cell infiltration, associated with activation of the STING‐IRF3 pathway and local immunogenic cell death.

Towards Robust Variational Quantum Simulation of Lindblad Dynamics via Stochastic Magnus Expansion

open access: yesPRX Quantum
Dedicated to the centenary of quantum mechanics. In this paper, we introduce a novel and general framework for the variational quantum simulation of Lindblad equations.

USP29‐regulated noncanonical stabilization of the hypoxia‐inducible factor‐α in aggressive prostate cancer

open access: yesMolecular Oncology, EarlyView.
We identify USP29 as the only DUB mirroring CA9 expression, a marker of hypoxia and HIF pathway activation associated with PCA aggressiveness. USP29 stabilizes HIF‐1α and HIF‐2α via a noncanonical mechanism that is independent of PHD/pVHL activity yet relies on proteasomal regulation, establishing USP29 as a previously unrecognized regulator of hypoxic

Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Preserving Multiple Conserved Quantities of Stochastic Differential Equations via Projection Technique

open access: yesMathematics
Stochastic differential equations (SDEs) with multiple conserved quantities are ubiquitous in scientific fields, modeling systems from molecular dynamics to celestial mechanics.
