Fluorescent probes allow dynamic visualization of phosphoinositides in living cells (left), whereas mass spectrometry provides high‐sensitivity, isomer‐resolved quantitation (right). Their synergistic use captures complementary aspects of lipid signaling. This review illustrates how these approaches reveal the spatiotemporal regulation and quantitative

Antigen-Drug Conjugates as a Novel Therapeutic Class for the Treatment of Antigen-Specific Autoimmune Disorders [PDF]
Multiple sclerosis represents the world’s most common cause of neurological disability in young people and is attributed to a loss of immune tolerance toward proteins of the myelin sheath.

This study reveals how the mitochondrial protein Slm35 is regulated in Saccharomyces cerevisiae. The authors identify stress‐responsive DNA elements and two upstream open reading frames (uORFs) in the 5′ untranslated region of SLM35. One uORF restricts translation, and its mutation increases Slm35 protein levels and mitophagy.

Tau acetylation at K331 has limited impact on tau pathology in vivo
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

CLASSIFICATION OF PSEUDOMONAS AERUGINOSA O ANTIGENS BY IMMUNOELECTROPHORESIS [PDF]
Heated saline extracts of 89 strains, and (1) supernates of phenol-water extracts (L1 fractions), (2) purified lipopolysaccharide, (3) trichloracetic-acid (TCA) extracts, and (4) sodium-hydroxide extracts of 23 strains representing all Pseudomonas aeruginosa O antigens were subjected electrophoresis.

Calpain small subunit homodimerization is robust and calcium‐independent
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Cloned long-term cytolytic T-lymphocyte line with specificity for an immediate-early membrane antigen of murine cytomegalovirus [PDF]
Long-term cytolytic T-lymphocyte (CTL) lines that are specific for distinct antigens associated with different phases of the replicative cycle of the murine cytomegalovirus (MCMV) were established by cloning of CTL lines derived from lymph nodes of ...

Rejection of Experimental Hodgkins Lymphoma by T-cells Engineered with a CD19 Chimeric Antigen Receptor [PDF]
T cells engineered to express chimeric receptors combining an external antibody binding domain with the CD3ζ T cell receptor (TCR) internal domain for triggering cell activation are being used for immunotherapeutic targeting of tumour cells in a non-HLA ...

The role of the O antigen in adjuvant activity of lipopolysaccharide [PDF]
Adjuvant activities of isogenic Salmonella enterica, serovar Typhimurium, O-6,7 and O-4,5,12 lipopolysaccharide (LPS), lipid A and Bordetella pertussis LPS were compared by immunizing groups of mice subcutaneously with diphtheria and tetanus toxoid vaccine alone or mixed with one of the LPS derivatives.
