Generation of Amplifiable Genome-Specific Oligonucleotide Probes and Libraries

open access: yesBioTechniques, 2002
Here we describe a process for the generation of oligonucleotide libraries representative of a given nucleic acid. Starting from a random pool of DNA oligonucleotides, the technique selects only those that hybridize to the nucleic acid template.

Microfluidic cartridge with integrated array of amorphous silicon photosensors for chemiluminescence detection of viral DNA [PDF]

open access: yes, 2016
Portable and simple analytical devices based on microfluidics with chemiluminescence detection are particularly attractive for point-of-care applications, offering high detectability and specificity in a simple and miniaturized analytical format ...

Oligonucleotide microarray analysis of aminoallyl-labeled cDNA targets from linear RNA amplification

open access: yesBioTechniques, 2004
Single-stranded long oligonucleotide-based (50- to 70-mer) microarrays offer several advantages over conventional cDNA microarrays. These include the easy preparation of the probes, low cost of array production, and low cross-contamination during probe ...

Motif effects in Affymetrix GeneChips seriously affect probe intensities [PDF]

open access: yes, 2012
An Affymetrix GeneChip consists of an array of hundreds of thousands of probes (each a sequence of 25 bases) with the probe values being used to infer the extent to which genes are expressed in the biological material under investigation. In this article,

Microarray-Based Method for Combinatorial Library Sequence Mapping and Characterization

open access: yesBioTechniques, 2003
Here we describe a DNA-chip-based method for high-throughput sequence mapping. This involves competitive hybridization between short and differentially labeled fluorescent oligonucleotide probes and glass-supported PCR products.

Sequence determinants of RNA G‐quadruplex unfolding by Arg‐rich regions

open access: yesFEBS Letters, EarlyView.
We show that Arg‐rich peptides selectively unfold RNA G‐quadruplexes, but not RNA stem‐loops or DNA/RNA duplexes. This length‐dependent activity is inhibited by acidic residues and is conserved among SR and SR‐related proteins (SRSF1, SRSF3, SRSF9, U1‐70K, and U2AF1).

Molecular probe technology detects bacteria without culture

open access: yesBMC Microbiology, 2012
Background Our ultimate goal is to detect the entire human microbiome, in health and in disease, in a single reaction tube, and employing only commercially available reagents.

Adsorption models of hybridization and post-hybridisation behaviour on oligonucleotide microarrays

open access: yes, 2006
Analysis of data from an Affymetrix Latin Square spike-in experiment indicates that measured fluorescence intensities of features on an oligonucleotide microarray are related to spike-in RNA target concentrations via a hyperbolic response function ...

Probe selection for high-density oligonucleotide arrays [PDF]

open access: yesProceedings of the National Academy of Sciences, 2003
High-density oligonucleotide microarrays enable simultaneous monitoring of expression levels of tens of thousands of transcripts. For accurate detection and quantitation of transcripts in the presence of cellular mRNA, it is essential to design microarrays whose oligonucleotide probes produce hybridization intensities that accurately reflect the ...

Organ‐specific redox imbalances in spinal muscular atrophy mice are partially rescued by SMN antisense oligonucleotides

open access: yesFEBS Letters, EarlyView.
We identified a systemic, progressive loss of protein S‐glutathionylation—detected by nonreducing western blotting—alongside dysregulation of glutathione‐cycle enzymes in both neuronal and peripheral tissues of Taiwanese SMA mice. These alterations were partially rescued by SMN antisense oligonucleotide therapy, revealing persistent redox imbalance as ...
