Viral Dynamic Model‐Informed Dose Selection of Mosnodenvir Prophylaxis against DENV‐3 in a Controlled Human Infection Model

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
Dengue virus (DENV) infection is a significant global health threat. Mosnodenvir (JNJ‐64281802) is a pan‐serotype small molecule antiviral blocking DENV replication. A Phase IIa study (NCT05048875) evaluated the antiviral activity, safety, and pharmacokinetics of mosnodenvir as prophylaxis against DENV‐3 in a controlled human infection model (CHIM).

Triglyceride Polygenic Score Identifies Individuals Who May Respond Differently to Aspirin in Primary Prevention

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
Low‐dose aspirin is no longer routinely recommended for the primary prevention of cardiovascular disease in older adults due to a lack of net benefit over bleeding risk. We hypothesized that genetic subgroups may experience differential harm or benefit from aspirin therapy.

Model‐Informed Approaches for Pediatric Type 2 Diabetes Evidence Generation: Insights from a Model‐Based Longitudinal HbA1c Meta‐Analysis from EMA Marketing Authorization Applications

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
Evidence generation for antihyperglycemic therapies in pediatric type 2 diabetes mellitus (T2DM) remains challenging, with most randomized trials failing to demonstrate statistically significant reductions in glycated hemoglobin (HbA1c), partly due to substantial between‐subject variability.

Even when you know it is a placebo, you experience less sadness: First evidence from an experimental open-label placebo investigation

Journal of Affective Disorders, 2022
BACKGROUND Recent studies demonstrate substantial effects of deceptive placebo on experimentally induced sadness, even on autonomic activity. Whether deception is necessary, remains to be elucidated.
