Multi‐regional Organoid Biobank Reveals FAK‐ACSL1‐Driven Doxorubicin‐Resistance and Predictive Biomarkers in Breast Cancer

open access: yesAdvanced Science, EarlyView.
This study established a high‐quality organoid biobank derived from 68 tumor sites across 50 Chinese patients, elucidated the drug sensitivity‐based molecular subtyping in breast cancer, and revealed a novel mechanism of drug resistance mediated by the FAK‐ACSL1 pathway.

Microfluidic nanoparticle synthesis for oral solid dosage forms: A step toward clinical transition processes [PDF]

open access: yes
Nanomedicine provides various opportunities for addressing medical challenges associated with drug bioavailability, stability, and efficacy. In particular, oral nanoparticles (NPs) represent an alternative strategy to enhance the solubility and stability

Prednisone raw material characterization and formulation development

open access: yesBrazilian Journal of Pharmaceutical Sciences, 2018
Solid dosage forms for oral use, particularly tablets, are the most highly used dosage forms in therapy because they are easily administered, have high productivity and relatively low cost and provide a more stable drug to form a semi-solid net. Numerous

Targeted β‐Glucan‐Veiled Oral Apremilast Nanotherapy Modulates Key Dysbiosis‐Associated Gut Microbiota and Alleviates Ulcerative Colitis‐Associated Anxiety, Depression, and Neuropsychiatric Behaviors

open access: yesAdvanced Science, EarlyView.
This study introduces β‐glucan‐shielded apremilast laden nanomicelles that withstand gastrointestinal degradation, selectively accumulate in inflamed colon tissue, and modulate the microbiota–gut–brain axis. The formulation restores barrier integrity, reduces systemic and neuroinflammation, and improves anxiety‐ and depression‐like behaviors in colitis

Probiotic‐Derived Vesicles Rectally Delivered via Thermo‐Gelling Copolymer Promote Mucosal Healing and Reduce Inflammation in Ulcerative Colitis

open access: yesAdvanced Science, EarlyView.
A thermo‐responsive rectal delivery platform integrating probiotic‐derived extracellular vesicles within a hyaluronic acid‐based copolymer enables localized and sustained therapy for ulcerative colitis. The formulation effectively suppresses inflammation, promotes macrophage polarization toward a regenerative phenotype, restores epithelial barrier ...

Hollow Cu2O Nanozymes Enhance Probiotic Therapy for Colitis via Redox Homeostasis and TXNIP/NLRP3 Inflammasome Inhibition

open access: yesAdvanced Science, EarlyView.
Hollow cuprous oxide (H‐Cu2O) nanozymes feature enlarged catalytic surfaces for superior reactive oxygen species (ROS) scavenging. By efficiently neutralizing mucosal ROS, H‐Cu2O directly suppresses the TXNIP/NLRP3 inflammasome axis and restores intestinal epithelial barrier integrity.

Targeted Degradation of Picornaviral 3C Protease via PROTACs Confers High Barrier to Viral Resistance and Broad‐Spectrum Antiviral Activity

open access: yesAdvanced Science, EarlyView.
This study reports D34 as the first PROTAC degrader that targets the 3C protease of picornaviruses. D34 effectively degrades EV71 3C protease via the ubiquitin‐proteasome pathway. More importantly, D34 exhibits a high resistance barrier and shows broad‐spectrum antiviral activity against multiple picornaviruses, highlighting its potential as a novel ...

Development of solid paediatric oral dosage forms

open access: yes, 2010
The paediatric population includes a category of patients with dynamic changes (due to the continuous body growth). Therefore their treatment requires individualized drug dosing (according to body weight) and the availability of different dosage forms ...
