Exponential Clonal Expansion of 5-Fluorocytosine–Resistant Candida tropicalis and New Insights into Underlying Molecular Mechanisms [PDF]

open access: yesEmerging Infectious Diseases
In 2022, we initiated systematic 5-fluorocytosine susceptibility testing of Candida spp. isolates in Denmark; we observed a bimodal MIC distribution in C. tropicalis, with MICs >16 mg/L in half the isolates.

Analyzing the Reaction of Orotidine 5'-Phosphate Decarboxylase as a Way to Examine Some Key Catalytic Proposals. [PDF]

open access: yesJ Am Chem Soc, 2023
This study analyzes the origin of enzyme catalysis by focusing on the reaction of orotidine 5'-phosphate decarboxylase (ODCase). This reaction involves an enormous catalytic effect of 23 kcal/mol that has been attributed to reactant state destabilization associated with the use of binding energy through the so-called Circe effect.

Crystallization of native and selenomethionyl yeast orotidine 5′-phosphate decarboxylase [PDF]

open access: yesActa Crystallographica Section D Biological Crystallography, 2000
Crystals of the Saccharomyces cerevisiae pyrimidine biosynthetic enzyme orotidine 5'-phosphate decarboxylase (ODCase) were grown by the hanging-drop vapor-diffusion technique at 277 K using polyethylene glycol 4000 as the precipitant.

Establishment of a selectable marker recycling system for iterative gene editing in Fusarium fujikuroi [PDF]

open access: yesSynthetic and Systems Biotechnology
Gibberellic acid (GA3) is a vital plant growth hormone widely used in agriculture. Currently, GA3 production relies on liquid fermentation by the filamentous fungus Fusarium fujikuroi.

Indiscriminate binding by orotidine 5'-phosphate decarboxylase of uridine 5'-phosphate derivatives with bulky anionic c6 substituents. [PDF]

open access: yesBiochemistry, 2007
Orotidine 5'-phosphate (OMP) decarboxylase appears to act upon its substrate without the intervention of metals or other cofactors and without the formation of covalent bonds between the enzyme and the substrate. Crystallographic information indicates that substrate binding forces the substrate's scissile carboxylate group into the neighborhood of ...

Increased activities of hepatic orotidine 5′‐phosphate pyrophosphorylase and orotidine 5′‐phosphate decarboxylase induced by orotate [PDF]

open access: yesFEBS Letters, 1972
Administration of orotate, an intermediate in pyrimidine nucleotide biosynthesis, has been shown to elevate the levels of acid soluble uracil [l-6] and cytosine nucleotides [S] in liver. Furthermore, activities of enzymes involved in the de novo synthesis of pyrimidine nucleotides increase following the administration of orotate; this has been ...

Development and application of a fast and efficient CRISPR-based genetic toolkit in Bacillus amyloliquefaciens LB1ba02

open access: yesMicrobial Cell Factories, 2022
Background Bacillus amyloliquefaciens is generally recognized as food safe (GRAS) microbial host and important enzyme-producing strain in the industry. B.amyloliquefaciens LB1ba02 is a production strain suitable for secreting mesophilic α-amylase in the ...

Orotidine-5′-Phosphate Decarboxylase and Pyrophosphorylase of Bean Leaves [PDF]

open access: yesPlant Physiology, 1967
This report includes results demonstrating the existence of orotidine-5'-phosphate decarboxylase and orotidine-5'-phosphate pyrophosphorylase in plant leaves. The decarboxylase enzyme, purified 8 fold from leaves of etiolated pinto beans (Phaseolus vulgaris L.), had a pH optimum of 6.3.

Mild orotic aciduria in UMPS heterozygotes: a metabolic finding without clinical consequences [PDF]

open access: yes, 2017
BACKGROUND: Elevated urinary excretion of orotic acid is associated with treatable disorders of the urea cycle and pyrimidine metabolism. Establishing the correct and timely diagnosis in a patient with orotic aciduria is key to effective treatment ...
