APOBEC mutagenesis, kataegis, chromothripsis in EGFR-mutant osimertinib-resistant lung adenocarcinomas. [PDF]

open access: yes, 2022
BACKGROUND Studies of targeted therapy resistance in lung cancer have primarily focused on single gene alterations. Based on prior work implicating APOBEC mutagenesis in histological transformation of EGFR-mutant lung cancers, we hypothesized that ...

Determining the optimal approach to identify osimertinib resistance; the first line osimertinib cohort of the OSIRIS study

open access: yesLung Cancer
Resistance mechanisms (RMs) inevitably develop during osimertinib treatment in EGFR mutation positive (EGFRm+) NSCLC, and may guide subsequent therapy. We hypothesized that a complete RM analysis requires next-generation sequencing (NGS) of tumor tissue and plasma in patients experiencing disease progression (PD) on first-line osimertinib. At time of PD

LINC01605 Predicts the Poor Prognosis of Non‐Small Cell Lung Cancer and Promotes Chemotherapy Resistance by Regulating miR‐7111‐5p/ELK1

open access: yesThe Kaohsiung Journal of Medical Sciences, EarlyView.
ABSTRACT Non‐small cell lung cancer (NSCLC) is characterized by high incidence, mortality, and poor patient prognosis, with chemotherapy resistance being a common challenge. This research intends to identify key molecules involved in NSCLC and elucidate the regulatory role of the LINC01605/miR‐7111‐5p/ELK1 axis in chemotherapy resistance. LINC01605 and

Restoration of Osimertinib sensitivity in lung cancer through BRD4 inhibitor-mediated depalmitoylation of mutant EGFR via APT1

open access: yesnpj Precision Oncology
Non-small cell lung cancer (NSCLC) with epidermal growth factor receptor (EGFR) mutations initially responds to the third-generation EGFR-tyrosine kinase inhibitor (TKI) Osimertinib.

Comprehensive genomic profiling of mucosal melanoma reveals novel fusion transcripts and dysregulation of cell‐cycle, MAPK, and PI3K pathways

open access: yesThe Journal of Pathology, EarlyView.
Abstract Mucosal melanomas (MMs) are rare, aggressive cancers with poor outcomes and limited response to standard therapies. A significant knowledge gap exists regarding their genomic landscape and corresponding druggable targets. This study explores this issue through a multi‐omic analysis, including whole exome‐, RNA‐, and targeted sequencing, of ...

Magnetothermal‐responsive drug release bone cement based on EGFR‐mutated lung cancer bone metastases

open access: yesVIEW, EarlyView.
A novel magnetothermal‐responsive bone cement was developed by integrating PMMA/CPC/Osimertinib matrix with Fe3O4@SiO2@Zol nanomaterials. This dual‐drug system achieves controlled release, combining excellent mechanical properties with potent anti‐tumor activity and bone regulation for treating EGFR‐mutated LC‐BoM.
