Inhibition of higher plant 2,3-oxidosqualene cyclases by nitrogen-containing oxidosqualene analogues

Phytochemistry, 1996
Abstract 10-Aza-10,11-dihydro-2,3-oxidosqualene (10 N-OS), 19-aza-18,19,22,23-tetrahydro-2,3-oxidosqualene (19 N-OS) and 23-aza-22,23-dihydro-2,3-oxidosqualene (23 N-OS), designed as analogues of carbocationic intermediates involved in the reaction pathway of 2,3-oxidosqualene-cycloartenol cyclase (OSCC) and/or 2,3-oxidosqualene-β-amyrin cyclase ...

Vinyl sulfide derivatives of truncated oxidosqualene as selective inhibitors of oxidosqualene and squalene‐hopene cyclases

Lipids, 2001
AbstractVarious vinyl sulfide and ketene dithioacetal derivatives of truncated 2,3‐oxidosqualene were developed. These compounds, having the reactive functions at positions C‐2, C‐15 and C‐19 of the squalene skeleton, were studied as inhibitors of pig liver and Saccharomyces cerevisiae oxidosqualene cyclases (OSC) (EC 5.4.99.7) and of Alicyclobacillus ...

Overexpression of functional human oxidosqualene cyclase in Escherichia coli

Protein Expression and Purification, 2015
The generation of multicyclic scaffolds from linear oxidosqualene by enzymatic polycyclization catalysis constitutes a cornerstone in biology for the generation of bioactive compounds. Human oxidosqualene cyclase (hOSC) is a membrane-bound triterpene cyclase that catalyzes the formation of the tetracyclic steroidal backbone, a key step in cholesterol ...

[Oxidosqualene cyclases in triterpenoids biosynthesis: a review].

Sheng wu gong cheng xue bao = Chinese journal of biotechnology, 2022
Triterpenoids are one of the most diverse compounds in plant metabolites, and they have a wide variety of physiological activities and are of important economic value. Oxidosqualene cyclases catalyze the cyclization of 2, 3-oxidosqualene to generate different types of sterols and plant triterpenoids, which is of great significance to the structural ...

Inhibition of oxidosqualene cyclase by substituted quinolizidines

Bioorganic & Medicinal Chemistry Letters, 1994
Abstract Substituted hydroxyquinolizidines have been synthesized as conformationally restricted analogs of the type II high energy intermediate formed during the cyclization of 2,3-oxidosqualene to lanosterol. Compounds 4a and 4b were found to be potent inhibitors of rat liver oxidosqualene cyclase (OSC) with Kn values of 0.51 μM and 0.11 μM ...

Protein engineering of oxidosqualene-lanosterol cyclase into triterpene monocyclase

Organic & Biomolecular Chemistry, 2013
A computational modeling/protein engineering approach was applied to probe H234, C457, T509, Y510, and W587 within Saccharomyces cerevisiae oxidosqualene-lanosterol cyclase (ERG7), which spatially affects the C-10 cation of lanosterol formation. Substitution of Trp587 to aromatic residues supported the "aromatic hypothesis" that the π-electron-rich ...

Conversion of a Plant Oxidosqualene-Cycloartenol Synthase to an Oxidosqualene-Lanosterol Cyclase by Random Mutagenesis

Biochemistry, 2002
A random mutagenesis/in vivo selection approach was applied to generate and identify mutations that alter the product specificity of oxidosqualene-cycloartenol synthase (CAS) from Arabidopsis thaliana. This work complements previous studies of triterpene cyclase enzymes and was undertaken to provide knowledge of the frequency and locations at which ...

Soybean germ oil inhibits oxidosqualene cyclase in 3T3 fibroblasts

European Journal of Lipid Science and Technology, 2005
Soybean germ oil (SGO) could take an important place among nutraceuticals if the finding were confirmed that a daily intake of SGO helps to maintain a normal level of serum cholesterol. Adapting the rice milling technology to the soybean refining process, we achieved a rapid and efficient separation of soybean bran and cotyledon from the hypocotyle ...

Photoaffinity Labeling of Oxidosqualene Cyclase and Squalene Cyclase by a Benzophenone-Containing Inhibitor

Biochemistry, 1998
A new orally active oxidosqualene:lanosterol cyclase (OSLC) inhibitor (Ro48-8071; Morand, O. H. et al. (1997) J. Lipid Res. 38, 373-390) showed potent noncompetitive inhibition of bacterial squalene:hopene cyclase (SHC) from Alicyclobacillus acidocaldarius (IC50 = 9.0 nM, KI = 6.6 nM) and OSLC (IC50 = 40 nM, KI = 22 nM for homogeneous rat liver OSLC ...

Functional diversity of oxidosqualene cyclases in genus Oryza

New Phytologist
Summary Triterpene skeletons, catalyzing by 2,3‐oxidosqualene cyclases (OSCs), are essential for synthesis of steroids and triterpenoids. In japonica rice cultivars Zhonghua11, a total of 12 OsOSCs have been found. While the catalytic functions of OsOSC1, 3, 4, 9, and 10 remain unclear, the functions of the other OsOSCs have been well studied.
